生体内二核及び多核銅中心の機構解明に関連する二核及び四核銅錯体の合成

1) オキシヘモシアニン及びチロシナーゼ機能モデルとしての二核銅錯体の合成:立体障害として6位にメチル基を導入した6-Mehexpy配位子(6-Mehexpy=1,2-bis[2-(bis(6-methyl-2-pyridyl)me-6-pyridyl]ethane)の二核銅(I)錯体と酸素分子との反応により室温で安定なμ-η^2:η^2-peroxodicopper(II)を得た。このO_2錯体は、極めて安定であり、酸素分子を解離するためには、約80°Cまで温度を上げなければならなかった。即ち、可逆的な酸素の吸着を行うことは困難であった。一方、オキシヘモシアニンは室温で容易に酸素を可逆的に吸着する。オキシヘモシアニンによる酸素分子の可逆的吸着機構を解明するために、6-Mehexpy配位子のエチレン架橋にエーテル酸素を導入した6-Me-O-hexpy配位子を合成した。6-Me-O-hexpy配位子を用いて、銅-銅間の距離を伸ばした二核銅錯体[Cu_2(OAc)_2(6-Me-O-hexpy)](ClO_4)_2(1)の合成に成功した。1は塩化メチレン中でH_2O_2と反応して、μ-η^2:η^2-peroxodicopperer(II)錯体錯体(2)を生成した。2は6-Me-O-hexpyの二核銅(I)錯体と酸素分子との反応からも生成した。2は-50℃以下でのみ安定であり-40℃付近では、酸素分子を脱着して、二核銅(I)錯体を再生した。酸素分子の可逆的吸着が数回繰り返された。これは、明らかに銅-銅間の距離が6-Me-O-hexpy配位子により伸ばされて、μ-η^2:η^2-peroxodicopper(II)錯体の熱的安定性が低下したためと考えられる。即ち、2はオキシヘモシアニンの酸素低親和性モデル化合物であり、オキシヘモシアニンが銅-銅間の距離を伸ばすことによって、酸素親和性を下げ、酸素分子を放出する機構を再現した最初のモデル化合物である。
2) 多核銅錯体触媒による効率的なフェノール類の酸素酸化反応の開発:フェノールやカテコールのO_2-酸化反応は、銅(II)イオンによって触媒され、活性種は四核銅錯体であると考えられてきた。しかし、これを実証した例はない。生体内では、活性中心に4つの銅イオンを持つマルチ銅タンパク質がフェノールなどを含む様々な基質のO_2-酸化反応を触媒している。我々は、これらの四核銅錯体の効果を実証し、酸化反応の加速効果の理由を解明するために、二核及び四核銅錯体を合成した。それらの錯体の構造を決定するとともに、これらが触媒するフェノール類のO_2-酸化反応を速度論的に検討した。その結果、カテコールのO_2-酸化反応が四核銅錯体によって加速されることが示された。, 日本学術振興会, 科学研究費助成事業, 1997 -1998, 奨励研究(A), 同志社大学
