  • エレクトロスピニングによる骨再生用アパタイト被覆ポリ乳酸ナノファイバー足場の開発

    生体内で早期にアパタイトで被覆されるポリ乳酸(PLLA)ファイバースキャホールドの開発を行った. アパタイト析出を誘起させるためりん酸水素カルシウム(DCPA)粒子を準備した.PLLAとPolyethylene oxide (PEO)を溶解させた1,3-dioxolaneにDCPA粒子を添加することでPLLA/DCPA溶液を調製した.PEOは相分離によりファイバーの多孔質化を促すために添加した.PLLA/DCPA溶液をエレクトロスピニング装置により紡糸し,多孔質PLLA/DCPAファイバースキャホールドを作製した.比較のため,DCPA粒子を添加しない多孔質PLLAファイバースキャホールドおよび中実PLLA/DCPAファイバースキャホールドを準備した.アパタイト析出を評価するため,スキャホールドを擬似体液(SBF)に37℃環境下で3日間浸漬した.浸漬後,SEMによるスキャホールド形態の観察を行った.また,EDS,FTIRおよびXRDによりスキャホールド表面および析出物の分析を行った. SEM観察により,SBF浸漬後にDCPA粒子を添加した多孔質および中実ファイバーの表面が析出物で覆われていることを確認した.DCPA粒子を添加しない多孔質ファイバー表面には析出物は観察されなかった.元素分析により析出物からCaとPが検出され,FTIRおよびXRD分析により析出物はアパタイトであることが確認された.中実ファイバーにおいてもDCPA粒子添加によってアパタイト形成が誘起されたが,部分的にクラックや剥離が観察された.ファイバー断面のSEM観察により,多孔質構造とした場合にはアパタイトがファイバー内部にまで析出していることが確認された.これらより,多孔質構造とすることでPLLA/DCPAファイバーが早期かつ均一にアパタイトで被覆されることが確認された., 日本学術振興会, 科学研究費助成事業 基盤研究(C), 2017/04 -2020/03, 基盤研究(C), 同志社大学
  • Development of in-situ high speed manufacturing process of carbon fiber reinforced thermoplastics with the help of carbon nanotube grafted carbon fiber

    Carbon nanotube (CNT) grafting method at the growth temperature of relatively low (550-600 degree Celsius), at which tensile strength of carbon fiber was not degraded, was developed by using Ni as the catalyst and ethanol as the carbon source by catalytic chemical vapor deposition (CCVD) method. CNT grafted carbon fiber showed higher fiber/matrix interfacial share strength than as received carbon fiber.   • Development of PLLA porous nanofiber scaffold for tissue regeneration by using electrospinning method

    In this study, PLLA porous hollow fiber scaffold was developed by using electrospinning to maintain nutrient supply in cultured cartilage. Porous hollow fiber scaffold was fabricated by using the co-axial spinneret. PLLA/PEO solution as the outer solution was prepared by dissolving poly-L-lactic acid (PLLA) and polyethylene oxide (PEO) in 1,3-dioxolane. 40% ethanol was used as the inner solution. According to SEM observation, the fabricated fiber had porous hollow structure.   • Development of Piezoelectric Scaffold using Lead-Free Piezoelectric Material MgSiO_3 for bone regeneration

    MgSiO_3 (MSO) thin film was fabricated by RF magnetron sputtering to develop new scaffold to accelerate bone formation. Single-layer, double-layer and triple-layer MSO thin film were fabricated on Cu/Ti/Si (100) substrate by RF magnetron sputtering. According to X-ray diffraction analysis, crystal growth along (111) orientation was confirmed for all MSO thin films. The surface potential was induced due to spontaneous polarization, and it decreased with increasing the number of layer. The culture system was developed to culture bone cells on the MSO thin film with 4-point cyclic bending deformation. Alternative sinusoidal waveform of the electric potential was observed for all MSO thin films, when pulsating cyclic bending deformation was applied to the MSO thin film by 4-point cyclic bending deformation. The peak value increased with the number of layer. These results showed that generated MSO (111) thin film can be used as coating material to enhance bone formation. In order to investigate the effects of cyclic surface potential of BaTiO_3 (BTO) induced with its deformation on osteogenic differentiation of osteoblast-like cells in vitro, osteoblast-like cells were cultured on BTO with or without compressive deformation. Polarized BTO and non-polarized BTO were prepared as specimen. Cyclic compressive load was applied to the BTO by a fatigue testing machine. The surface potential of polarized BTO due to spontaneous polarization was almost same as that of non-polarized BTO. Alternative sinusoidal waveform of the surface potential was observed on the BTO with frequency of deformation. ALP activity of osteoblast-like cells cultured on polarized BTO with compressive deformation was highest, whereas there were no significant differences in ALP activity of osteoblast-like cells cultured on polarized BTO and non-polarized BTO without compressive deformation.     In this study, an implantable piezoelectric thin film by using a newly developed biocompatible material MgSiO_3 (MSO) for hybrid actuator and enzyme fuel cell battery was generated. We obtained below described results. (1) A multi layered MSO film was generated by RF magnetron sputtering apparatus. It has highly oriented crystal structure. (2) A monomorph type actuator for Bio-MEMS devices, which will be implanted in the human body, was fabricated. It was confirmed that this actuator could transport water and blood serum. (3) An enzyme cell battery by trapping the glucose was generated. But the power was not enough for operate the Bio-MEMS device.   • Development of three-dimensional polylactic acid-piezoelectric material scaffold

    A cultivation system was designed to evaluate piezoelectric effects of the BaTiO_3 (BTO) ceramics on bone marrow cells, and rat bone marrow cells seeded on surfaces of the BTO ceramics were cultured in culture medium. ALP activity on the charged BTO surface was slightly higher than that on the non-charged BTO surface. The amount of calcium on the charged BTO surface was also higher than that on the non-charged BTO surface.   • 人の動作の冗長性の映像解析によるオンライン抽出

    今年度も,昨年度に引き続き,中華鍋操作の基本動作を一例にとりあげ,技能動作習得過程に関する基礎的な分析を進めた.まず,動作を,マーカの軌道や主成分得点の時系列推移によって特徴付けしたが,これでは,教師のアドバイスと非熟練者の習得過程との関連については十分な考察は困難であった.そこで,中華鍋操作の基本動作が周期的な動作であることに注目して,動作の基本周期,基本周波数の寄与率,標準偏差,位相差の四つの評価指標を用いて動作の詳細な特徴付けと考察を行った.この結果,習得過程におけるアドバイスの影響を示すことができ,習熟度によってアドバイス内容に差異があること,非熟練者によってアドバイスの解釈が異なること,などがわかった.また,非熟練者の状態変化を適切に把握するためには,外から観察するだけでは不十分であり,非熟練者と教師のやり取りなど,その状態に至るまでの経緯に対する理解が求められることが示された.教師は,アドバイスとそれによる動作の変化をみながら,非熟練者の状態に関する理解を深め,それに応じて,その都度アドバイスを追加・変更し,非熟練者の動作を矯正していく.このことは,アドバイス付きの技能動作の習得過程は,非熟練者が動作のコツを理解し,習得していく過程であるだけではなく,その非熟練者の状態を,教師がインタラクションを通じて把握していく過程でもあることを示している.したがって,技能動作習得過程の支援のためには,非熟練者の状態に関する理解を容易にするような,教師とのインタラクションの設計が一つのポイントとなる., 日本学術振興会, 科学研究費助成事業 萌芽研究, 2006 -2008, 萌芽研究, 京都大学

  • 生体用超高分子量ポリエチレンの疲労・破壊機構解明による信頼性確保

    人工関節用超高分子量ポリエチレン(UHMWPE)において,フレーク状の磨耗紛を伴ったデラミネーション破壊が多数報告されている.デラミネーション破壊は,摺動表面下に生じる繰り返し応力によって,表面下で疲労き裂が進展し,層状のはく離が生じる現象であり,滅菌に用いられるγ線照射と,その後の酸化劣化反応によって助長されると考えられているが,その発生メカニズムについては明らかになっていない.また,酸化防止剤として知られているビタミンEを添加することによって,UHMWPEの酸化劣化を低減し,デラミネーション破壊を抑制しようとする試みもなされているが,その抑制機構は不明である.疲労摩耗現象は非常に複雑であるため,本研究では,疲労摩耗現象における損傷を微視的な材料の引張-引張疲労破壊現象ととらえ,UHMWPEの疲労き裂進展特性に及ぼす,γ線照射,酸化劣化やビタミンE添加の影響を検討し,これにより,人工関節用超高分子量ポリエチレンの信頼性を確保することを目的とした.フーリエ変換赤外吸収分光測定結果から,γ線照射を行った後酸化促進試験を行った試料では,表面が酸化していたのに対して,ビタミンE添加材では,酸化が抑制されていた.CT (Compact Tension)試験片を用いた疲労き裂進展試験の結果を,試験中の試験片の塑性変形による影響を除くため疲労J積分値ΔJを用いて整理した結果,処女材,ビタミンE添加材に比べ,γ線照射を行った後酸化促進試験を行った試料では,同じき裂進展速度に対してのΔJ値が低下していた.それに対して,ビタミンE添加材に対してγ線照射を行った後酸化促進試験を行った試料のき裂進展曲線は処女材,ビタミンE添加材とγ線照射を行った後酸化促進試験を行った試料の間に位置していたことからビタミンEを添加することによるΔJ値低下の軽減が見られた.つまり,ビタミンE添加は,超高分子量ポリエチレンのγ線照射と酸化劣化によるき裂進展抵抗の低下を抑制する働きがあることが,定量的に明らかになった., 日本学術振興会, 科学研究費助成事業 若手研究(B), 2005 -2006, 若手研究(B), 同志社大学
  • Development of Mechanical and Fatigue Testing System for Micro Elements

    In order to develop a reliable micro system in a service operation, much care must be taken into micro mechanical evaluation, i.e., mechanical properties of μm-sized microelements including fatigue behavior. However, the evaluation method has not yet been established. In this investigation, mechanical testing systems for microelements under tensile and bending loads have been developed, and thereby fatigue testing at 1-200 Hz is possible. Moreover, mechanical tests were conducted for SiN thin films of 1 μm and 3 μm thick, and the effects of sample size and loading mode were investigated : the tensile strength increased with a decrease in sample size. However, the elastic modulus was independent of sample size, although it is dependent on a deposition method. Bending strength is less dependent on the sample size compared with those under tensile loading. However, the bending strength was higher than the tensile strength, indicating that the strength of a microelement is highly dependent on the loading mode. Moreover, the influence of micro artificial defect and fatigue loading on an optical fiber was investigated. The tensile strength was extremely sensitive to an environment, and the tensile strength decreased by moisture containing even in a vacuum at 10^<-5> Pa : the strength decreased with an increase in a partial pressure of water molecule and with a decrease in loading rate under both monotonic and cyclic fatigue loading. The estimation method for evaluating the life under constant and cyclic fatigue loading was proposed.   • アクチュエータ内蔵マイクロエレメント特性評価試験片の開発と動的環境強度特性評価

    マイクロマシンの実用化は,機械としての信頼性を確保した上ではじめて成り立つものである.とくに,近年では,Si単結晶ウエハなどを用いたバルクマシーニングに加え,ポリシリコン薄膜などの薄膜も構造材料として用いられるようになってきているが,これらの機械的特性あるいは疲労破壊特性については,標準的試験法すら確立されていない.マイクロエレメントの強度特性評価を行う場合に最も困難な点は,試験片のチャッキングである.圧子により荷重を負荷する曲げ試験は,試験片のチャッキングの問題を回避することができるため従来から多く適用されているが,応力勾配が存在すること,試験片と圧子の接触部で破壊することなどの問題がある.一方,微小な荷重を精度よく負荷できる試験機を用いた引張試験を用いて,静電チャックにより試験片を保持し,機械的特性評価も進められつつあるが,静電チャックでは,チャック部分のすべりが生じるため,疲労破壊特性評価には適さない.そこで,本研究では,ポリシリコン薄膜を対象として,接着剤によりチャッキングを行い,繰返し荷重下での疲労試験が可能なマイクロエレメント特性評価試験片を開発し,これを用いた引張試験によりポリシリコン薄膜の機械的特性評価を行った.さらに,乾燥空気中,実験室空気中,水中において疲労試験を実施し,ポリシリコン薄膜は,引張変動荷重下で水の影響を受けて破壊することを明らかにした.また,結晶粒径や結晶方位分布が,ポリシリコン薄膜のヤング率に及ぼす影響を,ボロノイ分割により作成した有限要素モデルを用いて検討した.それぞれの結晶粒に対してランダムに結晶方位を与えた場合は,解析領域内に含まれる粒子数が増加するにつれ,異方性による影響を受けにくくなり,平面ひずみモデルにおいてはヤング率は170GPaあたりに収束するのに対して,3次元モデルでは,ヤング率は160GPa程度になることを示した., 日本学術振興会, 科学研究費助成事業 若手研究(B), 2003 -2004, 若手研究(B), 京都大学
  • Development of environment controlled micro mechanical testing system for thin films

    In order to develop a reliable micro machine and MEMS in a service operation, much care must be taken into micro mechanical evaluation, i.e., mechanical properties of μm-sized microelements including fatigue behavior and effects of loading mode, e.g. tensile and bending loading. However, the evaluation method has not been well established yet. In this study, a bending testing system based on a Nano Indenter with knife-edge type indenter and a piezo stage were developed and the bending tests have been conducted for poly-Si thin microelements with 3.5μm thick, 5μm wide and 10μm long. Fracture stress obtained by the bending test was 3.9 ± 0.5GPa, which was higher than the strength under uni-axial tensile loading. To evaluate the effect of loading mode on the mechanical properties of a thin microelement, the uni-axial tensile testing system has also been developed. The system consists of a micromechanical tensile testing machine actuated by a PZT driven positioning stage, a micro manipulation system, a non-contact strain measurement system with a help of a double field of view microscope. By using this testing system, tension-tension fatigue tests for poly-Si thin microelements with 3.5μm thick and 10μm wide can be carried out up to 10 Hz of stress cycle frequency in various environments ; in wet environment, in dry air and in laboratory air. Moreover to evaluate the influence of the crystalline orientation, the number of grains and the grain size distribution on the elastic modulus of poly-Si thin film, finite element models were developed based on the Monte Carlo method.   • Investigation into the Degradation Mechanisms of Advanced Metallic Materials based upon Localized Hydrogen Distribution Analyses

    Advanced metallic materials are sensitive to an environment and in order to clarify the degradation mechanisms and to improve the toughness of the metallic materials in a service environment, we must clarify the process of hydrogen entry as well as the hydrogen trap sites in the material. In this research project, a method to analyze the localized hydrogen distribution has been developed: the method is based upon a silver decoration technique. The investigation yields the following conclusions, i) An amount of silver precipitated on a sample is increased with an increase in a hydrogen content of the test sample, ii) The sensitivity of the method is increased with a decrease in the pH value of the decoration solution, iii) A small particle of silver on the sample surface can be analyzed by a kind of a Kelvin force microscope. iV) The sensitivity of the method is extremely improved by coating a kind of metallic thin film on the sample surface. The important thing is the correlation between the hydrogen content and the amount of the precipitated silver on the sample surface. For this purpose, the hydrogen was charged from the one side of the sample, and the hydrogen which passed through the sample was detected on the other side of the sample. This shows a good agreement between the permeated hydrogen current or the hydrogen content of the sample and the amount of precipitated silver on the sample surface. This means that the developed method is useful to analyze the hydrogen distribution in a quantitative manner. The investigation also includes the evaluation of the environmental strength of a high-strength steel: the crack growth rate is increased under a cathodically charged condition, and the fracture surface became transgranular. The analyses of the localized hydrogen distribution as well as the thermal desorption behavior of hydrogen show that the charged hydrogen is mainly absorbed within a grain, and this agrees with the fracture surface morphology.   • Development of testing system for evaluating the mechanical properties of thin microelement

However, the evaluation method has not been well established yet. In this study, the testing system of a thin microelement has been developed to evaluate the mechanical properties; the tensile strength, elastic modulus, fatigue strength and so on, and the evaluation methods for thin microelement are established. The system consists of a micromechanical tensile testing machine actuated by a PZT driven positioning stage, a micro manipulation system, a non-contact strain measurement system with a help of a double field of view microscope. The system is basically designed for poly-Si thin microelement with 3.5μm thick and 10μm wide. When conducting the uni-axial tensile test, the most important and difficult thing is to establish the method of gripping the specimen. In this study, a gripping method with glue is developed and successfully applied to the uni-axial tensile test without any slipping at the grip. Moreover the testing system is installed on an AFM stage and in situ observation of specimen surface is successfully done.   • 繊維/樹脂界面の動的環境強度特性評価と界面はく離疲労き裂進展のその場観察

    複合材料は「組合せ」であるが故に数多くの特徴を有し,単一素材では実現できない特性の可能性に富んでいる一方,必然的に異種材料間に界面が存在し,その特性が複合材料の機械的特性に大きな影響を与える.しかしながら,繊維/樹脂界面の機械的特性や破壊機構,特に実使用下において重要となる動的環境強度特性に関する知識は,高強度・高弾性繊維強化複合材料の研究・開発の歴史が浅いこともあり,極めて乏しい状況にある.そこで,本研究においては,アラミド繊維とエポキシ樹脂を用いて,界面き裂を定量的に観察することができるモデルコンポジット試験片を考案し,乾燥空気中,実験室空気中,湿潤環境中において単繊維引き抜き試験を疲労荷重下で行い,界面はく離進展挙動に及ぼす水環境や繰り返し速度の影響を検討した.初期き裂の大きさは吸水時間に依存し,4週間吸水材は空中放置材に比べて大きな初期き裂が発生し,7週間吸水材の場合は埋め込み長さ全長にわたる初期き裂が生じた.初期き裂導入後のき裂進展については,界面き裂進展速度は界面き裂の進展にともなって低下し,最終的に停留した.界面き裂の停留は,最大荷重を界面き裂の表面積で除した値が,環境に固有な一定値に近づくことで条件付けられた.界面き裂は,乾燥空気中では実験室空気中に比べて高速に進展するのに対して,湿潤空気中ではほとんど進展しなかった.また,湿潤空気中でき裂進展試験を開始し,界面き裂が停留した後に乾燥空気を導入すると,き裂はふたたび進展を開始した.逆に,乾燥空気中で試験を開始し,界面き裂進展中に湿潤空気を導入すると,界面き裂は停留した.つまり,湿潤環境により,き裂進展速度が低下することがわかる.また,界面き裂進展速度は応力繰り返し速度に大きく影響を受け,繰り返し速度が低いときに速くなった., 日本学術振興会, 科学研究費助成事業 若手研究(B), 2001 -2002, 若手研究(B), 京都大学
  • Strength Evaluation of Single Crystal Silicon Microelements and Related Micromaterials

    In order to develop a reliable micromachine in a service operation, much care must be taken to micro mechanical evaluation, i.e., mechanical properties of μm-sized microelements. However, the evaluation method has not been well established. In this research project, the fracture behavior is investigated in the micromaterials for the micromachines and micro-electro mechanical system, including single crystal silicon. This will help us establish design criteria and life assessment methods for micromachines. The materials investigated were single-crystal silicon, poly-silicon film, and high-purity vitreous silica fiber. In this investigation, micro-mechanical testing machine is newly developed, and tensile testing of micro samples is thereby possible. The tensile strength of the poly-silicon films is about 0.9 GPa for the width of 10 μm, whereas the strength of the microsample with 5 μm is about 3.7 GPa, which are very strong compared with the structural materials for the ordinary sized machines and structures. Single-crystal silicon is immune to fatigue fracture and stress corrosion in laboratory air. However, the fracture behavior is sensitive to the water environment, and the synergistic effects of water environment and the dynamic stress caused a decrease in strength. Nanoscopic observation of the crack initiation site shows that the nanometric crack is formed on the surface owing to water and the dynamic loading, and the crack nucleation mechanisms are discussed. On the other hand, the influence of water environments on the fracture behavior is investigated in a high-purity vitreous silica fiber. Silica fiber is reported to be sensitive to water, and the static strength is decreases when the tests are conducted in water.   • Development of in situ manufacturing and mechanical evaluation system for micromaterials

    In order to develop a reliable micromachine in a service operation, much care must be taken to micro mechanical evaluation, i.e., mechanical properties of μm-sized microelements including fatigue behavior. However, the evaluation method has not been well established yet. In this study, the evaluation system of metallic foil is developed, including in situ specimen machining, chucking and non-contact strain measurement. Using an electric discharge machine (PX-05 : Mitsubishi Co.Ltd.), micro specimens of stainless steel foil were machined and a suitable machining condition was found to minimize the roughness of machined surface : the minimum of the maximum roughness was 1.0μm. The micro specimen for tensile testing was designed to make easily handle and testing, which has the specimen supporting part and the specimen chucking part. To measure the strain of microelements, for which strain gages on the market are too large and strong, a laser speckle strain measurement method was applied, and non-contact strain measurement system for microelements was developed. Integrating these machining system and non-contact strain measurement system, tensile and fatigue tests of micro specimens of stainless steel foil (thickness : 50 μm, width : 1 mm) was successfully conducted, and the influence of anisotropy and machining method on the mechanical and fatigue strength was investigated.   • Environmental Strength Evaluation and Investigation into Degradation Mechanisms of Advanced Metallic Materials based upon Analyses of State of Hydrogen and Nanofractography

    Advanced metallic materials are sensitive to an environment and in order to clarify the degradation mechanisms and to improve the toughness of these metallic materials in a service environment, we must clarify the process of hydrogen entry as well as the hydrogen trap sites in the material. In this research project, thermal desorption spectroscopy of hydrogens is applied to analyze the state of hydrogen in the material, or hydrogen trap sites, and correlate the responsible hydrogen in the material to hydrogen embrittlement : the materials investigated are titanium aluminides intermetallic compound and Ti-6Al-4V alloys, and the influence of environment on fatigue crack growth behavior was investigated. In the case of titanium aluminides, the tensile strength of the duplex material decreases in order of a water molecule content in an environment : the strength in vacuum is the highest, and decreases in order of laboratory air and water. In the case of the lamellar material, the fatigue crack growth rate in dry air is higher in the R-C crack plane orientation than that in L-C crack plane orientation : the crack growth rate becomes higher when the crack grows as the lamellae are tearing off. However, in the case of the duplex material, the crack growth rate in the R-C crack plane orientation is smaller in low ΔK (ΔK_) region. When the cathodic charging is applied, the fatigue crack growth rate becomes higher than that in dry air, in particular at higher stress intensity factor range. The hydrogen evolution rate is increased by cathodic charging, with lower temperature peaks and higher ones. The peaks at lower temperatures are correlated with hydrides decomposition and detrapping of hydrogen from microstructural imperfections such as microvoids. As received materials also shows a evolution peak at a higher temperature, and the evolution rate is almost independent of cathodic charging. In addition, the evolution rate at a higher temperature above 800℃ is increased by cathodic charging. These hydrogens are considered to have an important role on fatigue crack growth acceleration. In contrast with these, the crack growth rate of Ti-6Al-4V alloy is sensitive to environment when the crack plane orientation is T-L, whereas the crack growth rate in a solution is almost equal to that conducted in dry air. However, the crack path changes towards the longitudinal direction, when the applied stress intensity factor range exceeds a certain value.   • Development of Genetic Algorithms based System for High-Precision Reconstructions of Three-Dimensional Topographies using Stereo Fractographs

    A software is developed which enables reconstruction of the three-dimensional (3-D) shape of fracture surfaces without human assistance. It is based upon computer image processing and pattern recognition techniques by using a stereo-pair of scanning electron micrographs. The processing consists of two subprocesses : searching the matching points between two images and computation of heights using the relative shift of the matching points. By using the previously developed system, some mismatches were inevitable in the search process, in particular, for low-contrast SEM images such as striations, intergranular facets, and so on. In order to improve the accuracy of the search, a Genetic Algorithm (GA) was implemented into the developed system. By using the GA method, the 3-D shapes of a wide variety of fracture surfaces including cleavage failures, intergranular cracking, dimples and fatigue striations, were successfully reconstructed with sufficient accuracy. The searching processes by the GA method and the previously developed two-step algorithm of coarse and close searching were compared. These proved that the GA method has the advantage of accuracy in the searching process and of short-run-time. A detailed 3-D shape, or more than a 120×120 reconstructed point-sized shape, was thus obtained with sufficient accuracy and with a relatively short-run-time.
