  • Synthesis and utilization of ruthenium complexes with polystyrene-supported arene ligand

    In this study, the author investigated the synthesis of [(arene) RuCl2]2 having polymer-bound arene ligand, the complexation of small molecular ligand with it and its application to the catalytic reactions. When the reaction of polymer P-1 with [(ethyl 4-methylbenzencarboxylate)RuCl2]2 was performed in a mixed solvent of cyclohexane and chlorobenzene to afford dark green powder. 1H NMR analysis of the resulting powder implied the possibility of the formation of proposed ruthenium complex P-l-Ru. Treatment of P-l-Ru with a small molecular phosphine ligand Dppe (diphenylphosphinoethane) afforded yellowish powder.   • 金属触媒を用いるβ-水素脱離を制御したオレフィンへの求核付加反応の開発

    [RuCl_2(p-cymene)]_2をAgOTfで処理して得られる錯体にホスフィン配位子を組み合わせると,クロロホルム中で本反応に対する触媒活性を示すことが明らかとなった。スルホンアミド類とスチレンの反応においては二座ホスフィン配位子の利用が効果的であり,80%を超える収率で目的物を与えた。 ルテニウム前駆体に1,2-ビスジフェニルホスフィノベンゼン(DPPBen)を作用させた後,AgOTfで処理して得られた錯体の^1H NMRからp-シメンがルテニウム上に芳香環として配位していること,^<19>F NMRからルテニウムに配位したOTfと配位していないOTf^-が存在すること,^<31>P NMRからジホスフィン配位子がキレート配位していることが明らかとなった。また,単結晶X線構造解析にも成功し,NMRの解析結果が正しいこと,並びに,ルテニウムに配位したOTfは単座型であることが示された。単離・構造決定したルテニウム-DPPBen錯体を用いてスチレンに対する各種ヘテロ求核剤の付加反応を検討したところ,アルコールやカルボン酸さらにはスルホンアミドがスチレンに効率よく付加することを見出した。 Xantphos-ルテニウムカチオン性錯体は良好な触媒活性を示し,これまで反応の進行がほとんど見られなかったオレフィン類に対しても付加反応が進行することが示された。2-フェニル安息香酸のオレフィンに対する付加反応では,オレフィンとして1-ヘキセンや1-オクテンのような直鎖アルケン,(Z)-二置換であるシクロヘキセンでも80%を超える収率で反応が進行した。さらには,これまで反応しなかった脂肪族カルボン酸でも付加が観察され,マンデル酸が芦アリルアニソールに付加し,64%収率でマンデル酸3-(4-メトキシフェニル)-2-プロピルを与えた。, 日本学術振興会, 科学研究費助成事業, 2004 -2005, 特定領域研究, 同志社大学
  • Synthesis of α-Amino Acids via Phosphine-Catalyzed Three-Component Coupling Reactions

    Protonation of the highly reactive zwitterionic intermediate produced in the reaction of triphenylphosphine with dimethyl acetylenedicarboxylate from the protic substrate having carbonyl moieties leads smoothly to vinylphosphonium salt, which underwent an intramolecular Wittig reaction to afford heterocyclic compound in one-pot. This reaction was successfully applied to the intermolecular three-component coupling reaction of acetylenecarboxylates, protic nucleophiles, and aldehydes catalyzed by phosphine. It was found that three-component coupling reaction of phthalimide, ethyl propiolate, and aldehydes in the presence of a catalytic amount of triphenylphosphine proceeded to afford the corresponding γ-oxo-α-amino acid derivatives in moderate to good yields. An efficient synthesis of α,β-diamino acid derivatives via vinylphosphonium salts generated from ethyl propiolate and triphenylphosphine was also developed. That is the reaction of ethyl propiolate with phthalimide in the presence of a catalytic amount of tirphenylphosphine gave ethyl 2,3-diphthalimidoylpropanoate in almost quantitative yield in polar solvent.   • ホスホリン錯体の合成とその触媒作用の研究

    1.4,4',5,5'-テトラメチル-2,2'-ジホスホリン型配位子 Matheyらの報告に従い、上記ジホスホリン化合物の合成を行った。本化合物は、論文によると総収率10%程度で得られることが示されているが、我々の検討では、純粋なものは数パーセントしか得られないことがわかった。得られた化合物を用いてパラジウムとの錯体合成を行ったところ、二価のパラジウム化合物とは錯体を形成しなかった。このことは、本化合物が電子供与性が弱く、高原子価の金属錯体の安定化にはあまり効果がないことを示しており、我々の予測通りであった。0価のパラジウム錯体とは錯形成したと思われるスペクトルが得られたが、その充分なキャラクタリゼーションには至らなかった 2.フェナンスレンの1、10位がリンで置換されたジホスホリン型配位子の合成 一般に、1,10-フェナントロリンは、その強制的な平面構造のため、2,2'-ビピリジンよりも強い配位力をもつ。そこで、上記の2,2'-ジホスホリンよりもフェナンスレンの1、10位がリンで置換されたジホスホリン化合物がより配位子として有効であると考え、合成を試みた。合成経路として、2、3位を置換基で保護し、l、4位にリンと結合させるための官能基を持つアルキル基を導入したベンゼンを出発物質に用いる合成経路を検討した。側鎖の適当な位置にオレフィンを導入することでリン原子が導入できると考えられたので、2-メチル-3-プロペニルの導入を試みた。クロスカップリングの条件を検討することで、目的のジオレフィン化合物の合成に成功した。ついで、Friedel-Craft条件でのリン原子の導入に成功した。現在、熱もしくは光による環化反応の検討を行っている。この反応系では、大量合成が可能なことから、今後、反応条件の検討を行い、目的化合物を得て、錯体合成、触媒反応を検討していく予定である。, 日本学術振興会, 科学研究費助成事業, 1997 -1997, 萌芽的研究, 同志社大学
  • 配位力を制御した新規キラル錯体の合成とその触媒作用の研究

    1.モノホスフィンモノイリド型配位子並びに金属錯体の合成と触媒反応への応用 ビスホスフィン配位子TolBINAPをブロモ酢酸エステル、ブロモアセトニトリルと反応させることによりモノホスホニウム塩が選択的に得られ、それをメチルリチウムで処理すると、対応する光学活性モノホスフィンモノイリド(YLIPHOS)が得られた。次にYLIPHOSを配位子としたRu、Rh、Pdの錯体合成を行った。本YLIPHOSはRh、Pdと8員環キレート錯体を形成する。このとき、イリド炭素が中心金属に配位した金属-イリド炭素結合が形成されており、その配位力は、置換基や中心金属によって影響を受ける。また、この金属-イリド炭素結合は、配位と解離の平衡状態にあると考えることができる。ところが、Ru金属を用いた場合、YLIPHOSは単座の配位子として金属に配位していると思われる。得られた錯体の触媒活性を検討したところ、Pdの錯体がGrignard試薬とブロモベンゼンやアリルフェニルエーテルとのクロスカップリング反応の触媒として有効であり、光学収率6.5%並びに17%で生成物が得られた。この結果は、立体選択性は若干低いものの、本錯体が不斉触媒として機能することを示している。 2.アミノ酸を配位子に持つ新規不斉触媒の開発 アミノ酸は天然由来の安価なキラル化合物であるが、これまで後周期遷移金属錯体を用いる不斉触媒反応の配位子として有用な反応は見出されていない。本研究で、プロリンをルテニウムに配位させることにより、アセトフェノンがイソプロピルアルコールからの不斉水素移動反応に活性な触媒を生成することがわかった。室温24時間の反応で90%を越える収率と、79%の不斉収率で本反応が進行する。, 日本学術振興会, 科学研究費助成事業, 1996 -1996, 基盤研究(C), 同志社大学
  • Development of Novel Catalyst System for Highly Enantioselective Hydroformylations and Its Application for the Syntheses of Biologically Active Compounds.

    (1) Asymmetric hydroformylation of sulfur-containing olefins, such as vinyl-and allyl-sulfide, sulfoxide, sulfones has been performed in high enantioselectivities by using a rhodium complex of chiral phosphine-phosphite, (R,S) -BINAPHOS.The resulting oxo-aldehydes have potential use as synthetic intermediates for various biologically active compounds. (2) Asymmetric hydroformylation of conjugated dienes have also been examined. There are four carbons in the dienes which are capable to be attached to the formyl group. For various diences, the present system chooses one of the four possibilities, in high enantioselectivities. (3) Highly enantioselective copolymerization of propene and carbon monoxide has been performed by using Pd (II) - (R,S) -BINAPHOS.The catalyst system produces poly (alt-propene-CD) of the highest molecular weight as well as in the highest enantioselectivity ever reported.   • Development of Direct Synthesis of Optically Active Compounds

    Several BINAP derivatives have been prepared and their donor-acceptor propeties have been elucidated. New cationic complexes [RuX(binap)(arene)]Y were synthesized in high yields and their catalytic abilities as well as their behaviors in solution have been investigated. Optimization of the reaction conditions of asymmetric hydrogenation of methyl 2-benzamidomethy1-3-oxobutanoate by use of these complexes as catalysts afforded the corresponding alcohol in 98% de and 99% ee. Asymmetric hydrogenation of alpha-alkylidenecyclopentanones, alkylidenelactones, alkenyl ethers, and alpha-keto esters has also been performed in high enantioselectivities.   • 新規反応活性メタラサイクル化合物の合成、構造、反応性に関する研究

    環状有機化合物の環を構成する炭素の一部が金属で置き替った化合物はメタラサイクル化合物と呼ばれ、近年、遷移金属錯体を触媒とするメタセシス反応、カルベノイド反応、オリゴメリゼ-ション、重合反応など多くの重要な反応の中間体としてその化学が注目されている。本課題では主としてチタノセン残基を環内に含むチタナシクロペンタン及び1ーオキサー5ーチタナシクロペンタンの誘導体の合成、構造、反応性などについて研究を進めた。具体的には、本研究者らが既に錯体Cp^*_2Ti(C_2H_4)〔1〕(Cp^*はペンタメチルシクロペンタジエニル基)と高歪みチレフィンのメチレンシクロプロパン〔2〕とから初めて単離可能なほど安定なチタナシクロペンタン体〔3〕の合成に成功しているので、これらの成果をベ-スにしてさらに本分野の展開を計った。 (1)〔1〕と2ーフェニルメチレンシクロプロパン〔4〕の反応により得られるチタナシクロペンタン〔5〕の構造をX線構造解析により明らかにした。 (2)錯体〔1〕とフェニルアレン〔6〕ならびに2,2ージフェニルメチレンシクロプロパン〔7〕の反応を検討した。これらの歪化合物は、メチレンシクロプロパンの場合とは異なり、チタナシクロペンタンは得られず、〔6〕の場合は配位子交換により新規チタノセンーアレン錯体が得られ、〔7〕は1,1ージフェニルブタジエンに定量的に異性化した。 (3)錯体〔3、5〕の反応性を検討するために熱分解,加水分解、水素化分解を行った。熱分解反応では、βー炭素炭素結合開裂反応による生成物とともに、前周期メタラサイクル化合物では初めて還元的脱離による生成物が見られた。それらの化合物の生成経路を重水素標識実験により明らかにした。 (4)〔1〕と金属カルボニル化合物(Mn、Re、Cr、Mo、W)〔8〕の反応による親規オキサチタナシクロペンタンカルベンー金属錯体〔9〕の合成に成功した。錯体〔9〕の熱分解反応ならびに、アルキン類、イソニトリルとの反応を検討した結果、本錯体の構造上の特徴がその反応性に良く反映されていることがわかった。 (5)錯体〔5〕と一酸化炭素の反応を検討し、シクロペンタノン誘導体の生成を確認した。その反応機構を推定するためにオキサチタナシクロペンタン錯体〔10〕とCOの反応を行ったところ、チタン炭素結合にCOが挿入した6員環チタン錯体が得られ、X線構造解析によって本錯体がη^1ーアシル構造をもつことを初めて明らかにした。 (6)キラルなシクロペンタジエン化合物である1,5ージフェニルトリプタセペンの合成経路を確立し、チタン錯体の合成を行った。今後、この錯体の触媒活性などについてさらに検討を加える。, 日本学術振興会, 科学研究費助成事業, 1991 -1991, 一般研究(C), 京都大学
  • Development of New Chiral Transition-Metal Catalysts for Highly Efficient Asymmetric Synthesis

    We had already developed atroisomeric bis (triarylphosphine) ligand, 2, 2^'-bis (diphenylphosphino)-1, 1^'-binaphthyl (abbreviated to BINAP) and showed that its metal complexes had high efficiency as catalysts for asymmetric hydrogenation of -acylaminoacrylic acids and asymmetric isomerization of allylamines into enamines by 1, 3-hydrogen migration. We also reported synthesis of Ru (OCOR) _2 (binap), the first mononuclear BINAP-Ru (II) complexes, and demonstrated that they are highly efficient catalysts for the asymmetric hydrogenations of emanides, alpha, beta- and beta, gamma-unsaturated carboxylic acids, allylic and homoallylic alcohols. In order to expand the synthetic utility of BINAP ligand, we have continued our investigation on the development of new highly efficient asymmetric reactions catalyzed by various BINAP-Ru (II) complexes which have been summarized below. (1) New cationic BINAP-Ru (II) complexes [RuX (binap) (arene)]y (X=Cl, Br, and I ; Y = Cl, Br, I, BF_<4'> and BPh_4 ; arene = benzene and p-cymene) have been synthesized in high yields and the molecular structures have been determined by spectroscopic and X-ray analyses. We have shown that these complexes are highly efficient catalysts for enantioselective hydrogenation of various unsaturated substrates. Especially important is the high efficiency of these complexes as catalysts for asymmetric hydrogenation of functionalized ketones. (2) A number of substituted BINAP derivatives have been synthesized. Various factors controlling the catalytic activity and selectivity of the asymmetric hydrogenation of ketones have been elucidated by use of these complexes. (3) Highly diastereoselective hydrogenation of methyl 2-benzamidomethyl-3-oxobutanoate via dynamic kinetic resolution has been accomplished by using [RuI ((R-3, 5-di-tert-Bu-binap) (p-cymene)] I as catalyst, giving methyl (2S, 3R-2-benzamidomethyl-3-hydroxybutanoate (98% 99%), a versatile intermediate intermediate for the synthesis of beta-lactam antibiotics. (4) Mechanism of asymmetric hydrogenation of alpha,beta-unsaturated carboxylic acids catalyzed by Ru (OCOCH_3) _2 (bianp) has been elucidated based mainly on deuterium-labeled experiments. (5) New monocationic trinuclear complexes [Ru_3X_5 (binap) _3] Y have been prepared and their roles in catalytic reactions have been discussed.
