  • Possible mechanism underlying exercise training-mediated the differentiation of adipose-derived stem cells: roles of intracellular amino acid metabolism

    This study clarified the mechanism of exercise training (TR)-induced change in the differentiation ability of adipose-derived stem cells (ADSCs) to adipocytes by investigating amino acid-mediated signal transduction. TR inhibited adipogenic differentiation of ADSCs with reduced expression of PPARγ protein. TR also affected L-leucine-induced changes in the protein expression of Sestrin2 and mTORC1. However, these responses to TR differed between subcutaneous and visceral adipose tissue ADSCs. In particular, bioinformatics analysis revealed that the down-regulation of branched-chain amino acid degradation pathways was enriched in adipocytes differentiated from ADSCs derived from subcutaneous adipose tissue.   • Clarification of mechanism(s) underlying exercise training-induced possible change(s) in the differentiation potential of adipose tissue-derived stem cells

    Both visceral and subcutaneous adipose tissues are composed of several types of cells. Of these cells, adipose tissue-derived stem cells (ADSC) from either visceral or subcutaneous adipose tissue can be differentiated into each fat depot-specific adipocytes. The aim of the present project was to establish the effect of exercise training on the ability of ADSC into adipocytes and other type of cells. The main findings were as follows. First, fat depot-specific differences in developmental genes were found in the expressions, and some of developmental genes expressions were associated with exercise training-induced changes in adipose tissue weight. Second, exercise training affected intracellular metabolism, probably being associated with the changes in the ability of ADSC into mature adipocytes.   • The analysis for mechanism of exercise-induced adipose tissue remodeling on developmental genes

    Recent studies indicate that developmental genes are differentially expressed in subcutaneous adipose tissue (scWAT) and visceral adipose tissue (vWAT), and that some of these genes exhibit changes in expressions which closely correlate with the extent obesity and the pattern of fat distribution. The purpose of the current study was to investigate the effect of exercise training (TR) and high-fat diet (HFD) on developmental genes in three depots: inguinal, epididymal, and brown fat from rats. The fat depot-specific differences in mRNA levels of developmental genes were observed in three fat depots. TR and HFD induced the changes in the expression levels of some of these genes, i.e., HoxA5, HoxC9, and HoxC10. Moreover, the depression of HoxC10 in inguinal and brown adipocytes increases the expressions of brown fat genes, Ucp1 and Pgc1α.   • Response and adaptation of skeletal muscle to endurance training based on circadian rhythms of clock gene expression.

    The purpose of this study was to determine whether exercise timing enhanced exercise training (TR)-induced increase in the levels of mitochondrial proteins in skeletal muscles. The treadmill TR for 9 weeks was conducted at two time points (TR@zeitgeber time 0 (ZT0) and TR@ZT10). Both of per2 and pgc-1α mRNA expression showed the highest level at ZT0 and lowest level at ZT10. Additionally, the rats underwent acute exercise at the same time points (Ex@ZT0 and Ex@ZT10). The TR significantly increased levels of the mitochondrial protein (COXIV) in the deep portion of gastrocnemius muscle. Moreover, COXIV level in the TR@ZT10 group increased significantly compared to those in the TR@ZT0 group. Indeed, pgc-1α mRNA expression increased significantly immediately after acute exercise only at Ex@ZT10. The timing of TR influenced TR-induced adaptation.   • Establishment of the Evidence of Thermal Therapy before Exercise Protecting Articular Degeneration

    Cultured human articular chondrocytes were subjected to the 1st HS in a thermal bath (37, 39, 41, 43 °C for 20 min) after 48 h of differentiation. Then, 24 h after the 1st HS, half of them were exposed to fatal stress (2nd HS at 45 °C) for 15 min. Cell viability was measured 8 h after 2nd HS. Twenty-four hours after each HS, Hsp70, proteoglycan, and type II collagen mRNA expressions were quantified by rt-PCR. After the 1st HS, Hsp70 expression gradually increased with increase in temperature of the HS. Proteoglycan and type II collagen gradually increased until HS at 41 °C. HSP70 expression level significantly increased 24 h after the 1st HS at 41 °C compared with that in untreated chondrocytes. Moreover, chondrocytes pretreated at 41 °C showed increased cartilage matrix gene expression and the most tolerance to the 2nd HS.   • Exercise prescription based on the circadian rhythm of adipocyte clock genes : The significance of chrono-exercise prescription

    The mRNA levels of both adipokines and metabolic genes were strongly correlated with those of clock genes. The mRNA expressions of adipokines and metabolic genes, which were obtained after the exercise performed at the peak of Bmal1 mRNA expression, differed from those obtained after the exercise performed at the bottom of Bmal1 mRNA expression. Melatonin affected the mRNA expressions of several genes regulating adipocyte metabolism and adipokines.   • Optimal exercise intensity for defense ability to oxidative stress

    The purpose of the study was to evaluate the effects of physical exercise to antioxidation power in plasma. Six male subjects (21-25 years old) performed maximal exercise by a gradual increase load using a bicycle ergometer. Exercise load was raised by 25W every four minutes to exhaustion. Blood samples were obtained from the antecubital vein at rest, each load stage, and 10 minutes, 1-hour and 1-day after exercise. As antioxidation power, we measured iron reduction power (Fe^<3+>→Fe^<2+>). The quantity of hydroxyl radical generation was measured by a spin trap method. The lactic acid and the pyruvic acid were measured by an enzyme method. Six another subjects exercised with a physical work as equivalence. The iron reduction power in plasma rose with an increase in exercise intensity. On the contrary, the quantity of generation of a hydroxyl radical decreased with a rise of exercise intensity. This shows that antioxidation power of plasma increases by a rise of exercise intensity. In addition, there were negative relations to lactic acid and ESR signal intensity.   • キレート化合物による骨格筋遺伝子導入法の開発:スポーツ科学への応用

    われわれは、骨格筋細胞や組織に大きなダメージを与えることなく、遺伝子導入の効率を高めることができる遺伝子導入促進剤およびそれを含む遺伝子導入用組成物の国際特許取得のため、2004年9月10日に国際公開した。 すなわち、カルシウムイオンおよび/あるいはマグネシウムイオンを捕足する物質が、遺伝子導入効率を高めることを見出した。さらに、本研究は、遺伝子導入を望む外来遺伝子を含み、遺伝子導入される動物の骨格筋細胞中で外来遺伝子を発現することができる組み換えベクターと遺伝子導入促進剤とを媒体中に含む遺伝子導入用組成物を提供する。遺伝子導入促進剤は、カルシウムイオン/マグネシウムイオンを捕足する物質からなる。つまり、カルシウムイオン/マグネシウムイオンのキレート化剤が好ましい。たとえば、EDTA、CyDTA、DTPA-OH、EDDP、およびHDTA、ならびにこれらの塩(好ましいのは、ナトリウムやカリウムのようなアルカリ金属塩、複数のカルボキシル基の一部が塩になっているものを含有するもの)である。これらのうち、EDTAは細胞や組織へのダメージが軽微であり、医薬品としてすでに認可されているものでもあるので、最も望ましい。なお、カルシウムイオン/マグネシウムイオンを捕足する物質としてキレート剤は好ましいが、必ずしもキレート剤に限定されず、両イオンを捕足できる物質であれば他のものでもよい。 本研究により、遺伝子導入促進剤は、骨格筋細胞や組織に大きなダメージを与えることなく、遺伝子導入の効率を高めることができる。遺伝子導入促進剤の存在下に遺伝子導入を行うことにより、細胞や組織に大きなダメージを与えることなく細胞への遺伝子導入効率を向上させることができるので、本研究は、遺伝子治療の効率を高めることができ、遺伝子治療に有用な遺伝子導入用組成物を提供することが可能である。, 日本学術振興会, 科学研究費助成事業, 2003 -2004, Coinvestigator, 萌芽研究, 杏林大学
  • Clarification of the molecular mechanism(s) behind exercise training-induced suppression of fat mass

    In order to clarify the mechanism(s) behind either exercise- or exercise training-induced suppression of fat mass, the following studies were performed in rat adipocytes : (1)effects of exercise on apoptotic and anti-apoptotic signals, (2)effects of exercise and diet on several genes related with lipid metabolism, (3)effects of exercise on tumor necrosis factor-α (TNF-α)-induced apoptotic and anti-apoptotic signals, (4)effect of exercise training on insulin secretion from pancreatic islets, (5)alterations in exercise-induced β-adrenergic receptors and G proteins. As a result, the following findings were obtained. Exercise increased the Bcl2/Bax ratio in retroperitoneal and inguinal adipose tissue, but decreased its ratio in epididymal adipose tissue. Starvation reduced the expressions of some lipogenic genes, and re-feeding enhanced these expressions. Exercise after re-feeding blocked re-feeding-induced enhancement of some lipogenic genes expressions. Exercise training enhanced TNF-α signaling directed toward the expressions of survival signals and the suppression of fatty acid synthase gene expression. The mechanism behind exercise training-induced reduction of insulin secretion may involve an enhanced expression of neuronal nitric oxide synthase and a decreased expression of Gαi-2 protein. Acute exercise altered the expression of Giα2 protein via the ubiquitin-proteasome pathway, and the mechanism underlying the increased density of β_2-AR after exercise may involve alterations in a multi-step event involving the coordinate interaction among proteins mediating β_2-AR trafficking, e.g. G-protein-coupled receptor kinase-2 and β-arrestin-2.   • Possible role of nitric oxide in lipolysis in exercise-trained rats.

    A possible role of nitric oxide (NO) on adipocyte lipolysis was studied in exercise-trained (9 weeks of running) rats. Lipolysis in adipose tissue tended to be greater in trained rats than in control rats. Treatment of adipose tissue with 5 mM N^G -nitro-L-arginine methyl ester (L-NAME) showed that both basal and isoproterenol-stimulated lipolysis were significantly greater in trained rats than in control rats. In contrast, in isolated adipocytes, L-NAME had no effect on lipolysis in either group of rats, although the lipolysis of isolated adipocytes was significantly greater in trained rats than in control rats. Training significantly reduced nitrite/nitrate production in adipocytes but not in tissue. On the other hand, training increased the protein expression of endothelial nitric oxide synthase (eNOS) but not that of inducible NOS (iNOS) in the extracts of tissue homogenates. In tissue homogenates, eNOS activity but not iNOS activity was significantly greater in trained rats than in control rats. In cellular extracts, training significantly reduced the activites of both NOSs, but the mRNA expressions of both NOSs were not different between groups. The NO donor, S-nitroso-N-acetyl-penicillamine (SNAP), significantly inhibited adipocyte lipolysis in response to isoproterenol in both groups. This inhibitory effect of SNAP was greater in the adipocytes of trained rats than in those of the control rats. Thus, it is possible that NO is involved in the regulation of lipolysis and that exercise training enhances the responsiveness of adipocytes to extracellular NO with the reduced production of nitrite/nitrate in adipocytes due to decreased activites of NOSs.   • Possible role of adipocytokines in exercise-induced improvement of obesity and insulin-resistance.

    This research project examined possible role of adipocytokines in exercise-induced improvement of obesity and insulin-resistance. The increase in plasma concentrations of tumor necrosis factor (TNF)- α has been shown to result in insulin resistance. It is unclear, however, whether endurance exercise training can reduce the ability of adipocytes to secrete TNF- α. Therefore, we examined the effects of endurance training on the secretions of TNF- α and soluble TNF- α receptor (sTNFR) from isolated adipocytes. The expressions of menbrane-p55 TNF receptor (TNFR) in adipocytes were also examined. Male Wistar rats were divided into two groups, sedentary control and exercise-trained group. The rats of exercise-trained group were exercised on a rodent treadmill for 9 weeks. Adipocytes were isolated from the epididymal adipose tissue at least 36 hours after the last bout of exercise. The adipocytes (250000 per tube) were incubated in 5ml of DMEM/Ham's F12 medium for 24 hours at 37℃. The amounts of TNF- α and sTNFR in the culture medium were measured by ELISA.TNFR was measured by Western blot analysis. Exercise training increased significantly the secretions of TNF- α from isolated adipocytes. This increase was completely blocked by indomethacin or ETYA, cyclooxygenase inhibitors, suggesting that the enhanced secretions could be mediated by a possible change in the activity of cyclooxygenase. On the other hand, exercise training could decrease the secretions of sTNFR.The concentrations of sTNFR tended to be less in exercise-trained than in control rats. In contrast, exercise training could increase the expressions of TNFR proteins in adipocytes.     The effects of chronic physical exercise (ET) on the sensitivity of thoracic aorta to norepinephrine and on adipocyte lipolysis were investigated in spontaneously hypertensive rats (SHR). After ET, the attenuated sensitivity of aorta to norepinephrine was observed in SHR rats as well as in normotensive rats. This attenuated sensitivity of aorta to norepinephrine after ET was still evident in endothelium-denuded aorta, but to a lesser extent than in endothelium-intact aorta. No significant difference in 60 mM KC1-induced tension between control and trained SHR rats was observed with or without endothelium after ET.The results suggest that levels of endothelium-dependent releasing factor may increase in response to norepinephrine after ET in SHR rats as well as in normotensive rats. In the separate experiments, adipocyte lipolysis was investigated in SHR rats after ET.Adipocyte lipolysis stimulated by various agonists, such as isoproterenol, dobutamine, salbutamol, BRL37344 and dibutyryl cyclic AMP, was blunted in non-trained SHR rats compared with normotensive rats. However, ET significantly increased adipocyte lipolysis stimulated by each agonist, although the maximal responses were less in SHR rats than in normotensive exercise-trained rats.   • The role of Ca^<2+> and calmodulin in exercise trainining-enhanced lipolytic response.

    長期間の身体トレーニング(TR)は脂肪細胞の脂肪分解反応を増強させる.この現象は脂肪細胞のサイクリックAMP(cAMP)以降の酵素であるタンパクキナーゼ(PK)やホルモン感受性リパーゼ(HSL)の活性が増強するためであると考えられている.しかしながら,PKやHSL自身の活性はTRによって増強せず,未だその実体は捉えられていない.脂肪分解反応はcAMP以外にもCa^<2+>やカルモジュリン(CaM)によっても修飾されている.本研究においてはTRによる脂肪分解増強効果をCa^<2+>/CaM系とPKとの関係を検討した. TRによってラット脂肪細胞の脂肪分解反応は著明に増強した.TRラットおよびその対照群の脂肪分解反応はCaM阻害剤であるW-7で有意に抑制された.その抑制作用はTR群において有意に大きかった.このことからTRによる脂肪分解反応の増強機構にCa^<2+>/CaM系に大きく修飾されている可能性が示唆された.そこでさらにPK活性に及ぼすW-7の影響を検討した.細胞抽出液中のcAMPによるPK活性はTR群で低下する傾向にあった.このcAMPによるPK活性はW-7によって両群共に有意に抑制されたが,その抑制率はTR群(31.5%)で対照群(18.9%)に比較して有意に大きかった.このことから,TR群の脂肪細胞のcAMPによるPK活性の調節はCa^<2+>/CaM系に大きく依存していることが明らかになった.また,TRラットの脂肪細胞では細胞内Ca^<2+>濃度が有意に高く,これがCa^<2+>/CaMにより大きく修飾されているPK活性の調節に役だっている可能性も示唆された., Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for General Scientific Research (C), 1994 -1994, Principal investigator, Competitive research funding, Grant-in-Aid for General Scientific Research (C), The University of Electro-Communications
  • Effects of long-term physical exercise on contractile properties of rat aorta

    ラット大動脈の収縮特性に及ぼす長期間の身体運動(TR)の影響を内皮細胞との関連性から検討した.実験動物としてWistar系雄ラットを用い、TRは9週間のトレッドミル走行を実施した.TR終了後,ラットを屠殺し胸部大動脈を摘出後,種々の薬物に対する収縮特性を検討した.ノルエピネフリンによる収縮において,内皮細胞存在下でTR群の濃度反応曲線は対照群と比較し有意に右側にシフトした(50%有効濃度(ED_<50>),対照群;9.5nM,TR群;30nM).内皮細胞非存在下においても同様の現象が観察されたが,その差は小さくなった(対照群;ED_<50>=4.2nM,TR群;ED_<50>=9.7nM).外液Ca^<2+>のノルエピネフリン収縮に及ぼす効果にTRの影響は認められなかった.アンギオテンシンIIによる収縮は内皮細胞存在下でTRラットの濃度反応曲線は対照ラットと比較し有意に右側にシフトした(対照群;ED_<50>=2.1nM,TR群;ED_<50>=5.6nM).内皮細胞非存在下においても同様の現象が観察された(対照群;ED_<50>=2.0nM,TR群;ED_<50>=6.6nM).アセチルコリンによる弛緩やカリウム拘縮はTRによる影響を受けなかった.さらに急性の運動負荷について検討した.運動負荷後,対照群のノルエピネフリン収縮は内皮細胞の有無に関わらず濃度反応曲線は有意に右側にシフトした.TR群においても同様の現象が観察されたが,濃度反応曲線の右側へのシフトが小さく,結果的に運動負荷直後に測定したノルエピネフリン収縮は対照群とTR群で有意な差が認められなかった.また,運動負荷後,対照群のアンギオテンシンII収縮は内皮細胞の有無に関わらず濃度反応曲線は有意に右側にシフトした.一方,TR群のアンギオテンシンII収縮は急性の運動負荷の影響は認められなかった., Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Encouragement of Young Scientists (A), 1993 -1994, Principal investigator, Competitive research funding, Grant-in-Aid for Encouragement of Young Scientists (A), The University of Electro-Communications
