Selenium-transport mechanism of selenoprotein P

Selenoprotein P (SeP) is plasma selenoprotein containing essential trace element selenium. SeP functions as a transporter of selenium, delivering selenium to the cells. However, details of selenium-transport mechanisms of SeP remain unknown. In the present study, we identified SeP receptors in several tissues such as skeletal muscle, pancreas, and lymphocytes. Further, we identified SeP receptors with different affinity for SeP, and found a different molecular pathway to transport selenium of SeP to the cells. Development of diagnosis and treatment of Parkinson disease at early stage using novel biomarker oxidized DJ-1

DJ-1, the product of the causative gene of a familial form of Parkinson’s disease (PD), undergoes preferential oxidation at the cysteine residue at position 106 (Cys-106) under oxidative stress. In the present study using specific antibodies against Cys-106-oxidized DJ-1 (oxDJ-1), we evaluated oxDJ-1 levels in brain and blood of PD patients. It was found that DJ-1 oxidation in erythrocytes and in the brain occurs in PD patients, particularly during the early phases. DJ-1 oxidation was effectively inhibited by endogenous antioxidant glutathione. Activation of Arterial Wall Cells during Atherogenesis : A study using a in vitor co-culture model system

A new in vitro system for the co-culture of vascular endothelial cell(EC), smooth muscle cell(SMC) and monocyte/macrophage has been developed for the anaylysis of pathogenesis of atherosclerosis.
We have established a vascular cell under flow. Using this system we examined the molecular pathogenesis of atherosclerosis and endothelial activation.
Results
(1)Oxidative modification of lipoproteins : We established a method for tracing oxidative modification. Transcriptome analysis indicated lyso-PC affects many genes in EC.
(2)Using co-culture system, we found the involvement of NRF2 in shear stress sensing.
(3)Changes in transcriptome were analyzed during inflammation, growth and coagulation in EC. Adding to Ets, GATA and Hex, the role of NFATc in these signal transduction was found. We also found that DSCRl can inhibit NFATc signaling.
(4)We found a novel inhibitor for upregulation of adhesion molecule using co-culture system. To analyze in vitro the migration of monocytes to the subendothelial space, their differentiation into macrophages, and the subsequent formation of form cells in vitro, we developed a 2-coculture system with rabbit aortic endothelial cells, aortic smooth muscle cells, and a mixture of matrix proteins on polyethylene folters in chemotaxis chambers. When oxidized low density lipoprotein (LDL) was added to the matrixlayer of coculture, on day 4 transmigrant cells exhibited lipid deposit droplets, and by day 7, they had the appearance of typical foam cells.
To follow lipid oxidation of low density lipoprotein andwithin cells we developed a novel method using diphenyl-1-pyrenylphosphine (DPPP) as a fluorescent probe. DPPP reacted with organic hydroperoxides and hydrogen peroxide stoichiometrically to give DPPP oxide (DPPP=O). DPPP incorporated into phosphatidylcholine liposomal membranes and polymorphonuclear leukocytes (PMNs) reacted with methyl linoleate hydroperoxide rapidly but not with hydrogen peroxide nor with tert-butyl hydroperoxide. This novel method revealed that lipid peroxidation proceeded within membranes of PMN stimulated by phorbol-12-myristate-13-acetate, which is known to produce several kinds of free radicals. It was found that DPPP is a suitable fluorescent probe to monitor lipid peroxidation within the cell membranes specifically. DPPP incorporated into LDL gave a strong fluo-rescence within macrophage after macrophage took up DPPP-containing LDL.
We examined antioxidant activity of 4 isomers of vitamin E, (α- and γ-) tocopherol and tocotrienol, and inhibitory effect on expression of adhesion molecule of human endothelial cell. There were not significant deffereces in antioxidant activity and inhibitory effect on adhesion molecule expression. However, the concentrations of tocotrienols within cell were more than 10 times higher than that of tocopherol due to double bond in side chain of tocotrienol. By taking the defference in the concentrations of these antioxidants into considerations , tocopherols were more efficient in inhibition of the expression of adhesion molecules than tocotrienols. Development and Application of Quantitative Evaluation System for Oxidative Damage of Lipids

Lipids in oil products and foods are continuously damaged by aerobic oxygen and light. This results in quality deterioration and causes safety problems. Consequently, to inhibit such oxidation and to evaluate oxidative damage are of prime importance. The objective of this research is to develop a method by which the extent of oxidation of oils and lipids can be measured with high specificity, sensitivity, speed, and simplicity. It has been also aimed at developing and evaluating novel antioxidants which can effectively inhibit such oxidation. Lipid peroxidation products from fatty acids, phosphatidylcholines and cholesteryl esters were quantified regio-, stereo- and optical-specifically with a high performance liquid chromatography (HPLC) equipped with UV and chemiluminescence detector. The quantitative analyses have been successfully applied to oils, micelles, liposomes, lipoproteins, plasma and cells. This enabled us to distinguish and identify the active oxidants and mechanisms involved in lipid peroxidations. Furthermore, cholesterol esters were analyzed for different classes of fatty acids. Diphenylpyrenylphosphine (DPPP) was used to develop a ultra-sensitive analytical method. DPPP oxide formed stoichiometrically by the reaction of DPPP and lipid peroxides was analyzed and applied to the quantification of lipid peroxides. It was confirmed by this method that the location of lipid peroxides in the cells could be visualized with a conforcal fluorescence microscope. This method could be quite promising for further application and development. Measuring Lipid peroxidation in the matrix using ESR spin labeling

The novel method for measuring lopid peroxidation in the heterogeneous micro environments such as lopid bilayrs has been studied. This method might be useful for studying the degradation of newly synthesized materials such as functional polymers and biomembranes and also be useful for evaluating the activity of antioxidans which are designed to reside within the matrix.
Followings are the results obtained. First, we evaluated the degree of lipid peroxidation taking place in the artificial lipid bilayrs (liposome) prepared with phosphatidylcholine, using ESR spin label technique with 5,10 and 16NS as spin probes. When azo compound AAPH was used as a free radical generator, ESR signal intensity of spin probes were decreased with time-and dose-dependently. By comparing with the formation of lipid hydroperoxides this method was proved to be useful to evaluate lipid peroxidation of the artificial membranes quantiatively. We next tried to apply this method to biological materials. Human leukemia cell line THP-1 or U937 was labeled with spin probes 5NS and 16NS by incubating cells with thin film formed on the inner surface of a round-bottom flask. The various kinds of reactive oxygen species were applied. In the case of AAPH,an extracellular peroxyl radical generator, the signal intensity of 5NS was diminished faster than that of 16NS,suggesting that the amount of free radicals near the surface was greater than deep inside the membranes. On the other hand, hydrogen peroxide, one of the most common reactive oxygen species, did not reduce the signal intensities of these two spin probes significantly, suggesting that hydrogen peroxide itself has little interaction with the plasma membrane of the cell. Design and Evaluation of Novel Selenium-Containing Antioxidant

There is now increasing evidence which shows that the free-radical mediated oxidations proceed in vivo as in plastics rubber and lipids and that they play a causative role in the progression of various diseases, cancer and aging. As a consequence, the inhibition of such oxidations and development of novel potent antioxidants have received much attention. The present research has been carried out aiming at developing a novel selenium-containing antioxidant and evaluating its antioxidant potency. Ebselen, 2-phenyl-1,2-benzisoselenazol-3 (2H) -one, was chosen as a candidate and its antioxidant action and activity were studied. It was confirmed that ebselen did not act as a radical scavenger nor did it reduce vitamin E radical to regenerate vitamin E.Ebselen reduced hydroperoxides rapidly to corresponding alcohol and thereby inhibited the metal-catalyzed oxidations of lipids and low density lipoproteins. Furthermore, it was found that ebselen completely inhibited the lipid oxidation induce by lipoxygenase. Ebselen also exerted potent antioxidant activity against lipid peroxidation induced by peroxynitrite. リポ蛋白質の超分子構造と機能

LDLは,コレステロールエステル(CE)とトリグリセリド(TG)から成る核を,主としてホスファチジルコリン(PC)とコレステロールから成る一枚膜が外からとり囲んでおり,さらに分子量55万のアポタンパクB-100が全体をとりまくという,特徴ある超分子複合体である.ラジカルの攻撃を受け,酸化されるのは主としてPCとCEに含まれる高度不飽和脂肪酸部位である.興味深いことに,PCとCEの相対的酸化反応性が環境に大きく依存することを見出した.すなわちLDL中ではCEがPCよりも酸化されやすいこと、同じものでもt-ブチルアルコール中ではPCとCEが同程度酸化されやすくなり、一方ヘキサン中ではPCだけが圧倒的に酸化されることを認めた.
LDLの構造と反応性,その場での抗酸化物の作用機序を明かにするために,ドキシルステアリン酸をスピンプローブとしてLDL内にとりこませ,そのドキシル基の位置を変えることにより,LDL内の流動性,反応性を検討した.また,ドキシルステアリン酸のコレステロールエステルを合成し,コレステロールエステル転移蛋白(CETP)を用いてLDL内にとりこませ,同様の検討を行った.その結果,LDLの表層は流動性が低く,内部に入るほど流動性が高くなること,中心の核の流動性が大きいことが分かった.ドキシル基のニトロキシドはアスコルビン酸(ビタミンC)により速やかに還元されるが,LDLの内部に入るほど,水層のアスコルビン酸による還元速度が遅くなることを明らかにした.これはビタミンCとEによる相乗効果と関連して重要である.
LDLの酸化反応が連鎖的に進行し、CEとPCのヒドロペルオキシドを主生成物として与えることを確認した.アポ蛋白B-100の酸化に伴い、開裂による低分子化と橋かけ反応による高分子化が同時に進行するとともに、負負荷が増加していくことを確認した.そして重要なことは、それにつれてマクロファージによるとりこみも増加した., 日本学術振興会, 科学研究費助成事業, 1994 -1994, 重点領域研究, 東京大学
食品に含まれる抗酸化物質の機能解析と新規生体防御物質の設計

要旨:食品に含まれる種々の抗酸化物質およびそれらの類縁化合物、新規化合物の抗酸化作用について検討し、以下に示すような結果を得ている。
(1)カレー粉に含まれるカーキュミンをはじめコーヒー酸など数種の天然フェノール化合物の抗酸化活性を求めたが、α-トコフェロール(ビタミンE)を超えるものはなかった。
(2)β-カロテンをはじめとするカロテノイド類の抗酸化作用について基礎的検討を行ない、β-カロテンの抗酸化活性が、均一溶液中よりは膜中で、また酸素圧の低いほど抗酸化活性が大きいことを認めた。
(3)チオール化合物の抗酸化作用について研究をはじめた。
(4)トコフェロール、ヒドロキシカルバゾール、および2,6-ジ-tert-ブチル-4-ヒドロキシトルエン(BHT)など合成抗酸化物の膜における抗酸化作用について詳細に検討し、これらの相対的な活性や水相にあるアスコルビン酸との反応が、これらの膜内での存在位置、その場での動きやすさ、ラジカルの生成場に依存することを実験的に明らかにした。
(5)低比重リポタンパク質(LDL)の脂質の酸化、アポリポプロテインB100の変性に対する抗酸化物の抑制作用について検討し、不均一な場であるLDLの酸化に対しては、特に抗酸化物の存在場と動きやすさが重要な因子であることを明らかにした。

Development, Design and Evaluation of Novel Natural and Synthetic Antioxidants

This study has been carried out aiming at the development, design and evaluation of novel natural and synthetic antioxidants which effectively inhibit the oxidative deterioration of biological molecules, membranes and tissues as well as foods and oil products. Following results were obtained.
1. The antioxidant activities of vitamin E, phenolic compounds and carbazoles were evaluated quantitatively and it was shown that physical factors such as mobility at the microenvironment as well as chemical reactivities are quite important in determining overall antioxidant potency.
2. Inhibition of oxidation of low density lipoprotein, which is an important initial event of atherosclerosis, by antioxidant was studied in detail.
3. Inhibition of oxidation of lipids by various phenols, aminophenols and carbazoles was studied and the correlation between structure and chemical reactivity was discussed.
4. The kinetics of mechanisms of antioxidation by selenium containing novel compound was studied.
5. 食品に含まれる抗酸化物質の機能解析と新規生体防御物質の設計

フリーラジカルや活性酸素種により生ずる生体の傷害に対して、食品が有している防御因子を解明するとともに、その知見をもとによりすぐれた防御機能を有する物質を設計、開発することを目指して研究を進め、以下の結果を得た。
1)ビタミンE(トコフェロール)およびその類縁化合物の膜、低比重リポタンパク(LDL)中での抗酸化作用をスピンラベル法などにより検討し、ビタミンEの活性基が膜、LDLの表面に存在していること、動きが制限されていること、そのため内部のラジカルを捕捉しにくいことを明らかにした。
2)特にLDL中において側鎖の長さの影響が大きく、側鎖が短いほど抗酸化活性が大きくなることを見出した。LDL中では、抗酸化物の化学的反応性よりはむしろミクロな場での濃度、動きやすさがより重要な因子となることを明らかにし、そのためにビタミンEより化学的活性が小さいにもかかわらず、プロブコールがより強い抗酸化作用を持つことを示した。
3)放線菌Strep to myces chromotucusより抽出されたカラゾスタチンをはじめ種々のカルバゾール、アミノフェノール、クロマノール類の高度不飽和脂肪酸および脂質の酸化に対する抗酸化作用を定量的に検討した。
4)含セレン化合物であるエブセレンはラジカル捕捉能を有しないが、速やかにヒドロペンオキンドを分解するために、鉄あるいは銅により誘起される脂質の酸化反応を顕著に抑制することを明らかにした。, 日本学術振興会, 科学研究費助成事業, 1992 -1992, 重点領域研究, 東京大学
フリーラジカルによる細胞の酸化的傷害とその防御機構に関する基礎的研究

フリーラジカルが細胞、特に脳神経細胞に対して如何にして酸化的傷害を与えるのか、またそに対してどうすれば防御できるのかについて反応論的観点から基礎的に研究を行ない、解明することを目的として研究を進め、以下の結果を得た。潅流後、ラットより摘出した脳、肝のホモジネート(10mg protein/ml Tris-HCl緩衝液pH7.4)の酸化とその抑制について比較検討を行なった。ホモジネート中のビタミンE(α-トコフエロール)の濃度は、脳で0.12nmol/mg protein、肝では0.06nmol/mg proteinであった。EDTA存在下で水溶性ラディカル開始剤AAPH(2,2'-azobis(2-amidinopropane)dihydrochloride)10mM添加により、一定速度でペルオキシルラディカルを発生させて酸化を開始すると、肝では反応2時間までにビタミンEは消費され、3時間でTBARSは1.1nmol/mg protein増加した。同じ反応時間における脳のTBARSは0.3nmol/mg proteinでビタミンEの減少は10%以下であった。一方、EDTA非存在下で開始剤を添加しない場合、肝の酸化はほとんど起こらないのに対して、脳は速やに酸化し、0.05nmol/mg proteinのビタミンEが残存しているにもかかわらず、TBARSは1.48nmol/mg proteinに達した。以上の結果から、脳脂質は高濃度存在するビタミンEによってペルオキシルラディカルによる酸化はよく抑えられるが、金属依存性の酸化を受けやすく、またビタミンEはこれをよく抑制することができないことが示唆された。
