    • Mechanistic study on circadian clock system regulated by heme/CO crosstalk

      本研究では細胞内概日リズム制御におけるヘムと一酸化炭素のクロストーク性を解明することを目的としており,2021年度にはそれを可能とする化合物群の合成と,細胞内一酸化炭素を計測するための独自のアッセイを確立して報告した。まず細胞内一酸化炭素を捕捉するためのケミカルツールhemoCDを用いたアッセイを確立した。動物体内の細胞には微量の内因性一酸化炭素(CO)が常時存在するが,それを簡便かつ正確に計測する方法はこれまでに報告されていなかった。我々は細胞内や動物組織内に含まれるCOをhemoCDを使って簡便かつ正確に計測することを試み成功した。この定量方法により,デキサメタゾンで同期化した細胞内に含まれるCOの産生には概日リズムが存在することが明らかとなった。さらに動物を用いて組織内に含まれるCOの定量方法を確立した。この手法は,内在性COのシグナル伝達物質としての機能解明に役立つだけでなく,過剰のCOを吸入した体内における毒性メカニズム解明にも役立てることができ,論文公表後にプレスリリースにより広く周知した。その結果種々の共同研究および知財としての権利化を遂行することとなった。さらに概日リズム解明のためのケミカルツールとして,細胞内でCOを効率よく放出する新規CO-releasing moleculeの開発,ヘムオキシゲナーゼによって分解されてもCOを発生しないメソメチルヘム誘導体の合成にも成功しており,次年度からの研究に貢献するケミカルツールが種々揃っている段階である。, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Scientific Research (B), 2020/04 -2023/03, Grant-in-Aid for Scientific Research (B), Doshisha University
    • Development of innovative drugs having multistep action for treatment of central cerebral amyloidosis

      アルツハイマー病に対する薬剤の臨床試験は数多くなされているが、その成功例はほとんどなく、有効な治療薬の開発が強く望まれている。我々はアルツハイマー病に対する複数の危険因子に対して個々に作用する薬剤ではなく、5つの危険因子(脳におけるAPOε4遺伝子発現、コレステロール蓄積、脳炎症反応、アミロイド線維形成・蓄積、神経原繊維変化)に対して多段階かつ同時に抑制する薬剤の開発を期待して、我々が開発した新規化合物であるシクロデキストリン/ポリアミドアミンデンドリマー結合体(CDE)に、さらに静脈内投与後の有効性を可能にする血液脳関門(BBB)透過リガンドを結合したBPB-CDEの構築に関する研究を行っている。また、Apoε4遺伝子発現を特異的に抑制できるsiRNA(Apo-siRNA)とBPB-CDEとの複合体を調製し、それらの化学的・物理的性質、細胞膜透過性、Aβ凝集・繊維形成抑制・溶解効果などの検討を予定し、2021年度は次のような検討を行った。β-シクロデキストリンとPAMAMデンドリマーとの結合体(CDE)にBBB透過リガンドを結合体したBPP-CDEの合成・精製・構造解析を行った。Aβペプチド(1-40)及びAβペプチド(1-42)の凝集体形成に及ぼすPAMAMデンドリマー及びBPP-CDEの影響について検討を行った。また、Aβペプチド(1-40)とPAMAMデンドリマー及びBPP-CDE との相互作用をNMRを用いて検討した。PAMAMデンドリマー及びBPP-CDEとsiRNA複合体を調製し、その粒子径・PDI・ゼータ電位等の物性について測定する準備を行った。論文公表1件、学会発表2件を行った。, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Scientific Research (B), 2020/04 -2023/03, Grant-in-Aid for Scientific Research (B), Daiichi University, College of Pharmaceutical Sciences

      本研究では、肝臓の遺伝子疾患である家族性アミロイドポリニューロパチー(FAP)に対する根治療法としてのin vivoゲノム編集技術の開発を目的に、(1) 肝実質細胞特異的非ウイルスベクターであるデンドリマー/シクロデキストリン結合体(CDE)にラクトースまたはN-アセチルガラクトサミンおよびポリエチレングリコ―ル(PEG)を結合したそれぞれL-PaCおよびGN-PaCの構築、(2) ゲノム編集効率の向上を企図して、核内での遺伝子発現が不要なCRISPR mRNA及びガイドRNAの構築、(3) L-PaCまたはGN-PaCとCRISPR mRNA及びガイドRNAと複合体形成及びそれらの物理化学的性質の評価、(4) ヒト肝実質培養細胞(HepG2細胞)でのゲノム編集効率に関する基礎的検討を行うことを計画した。2019年度は、研究代表者の所属変更に伴う研究環境の大きな変化並びに新型コロナウイルス感染拡大に伴う研究制限等により、計画した研究を十分に遂行できなかった。このような状況から、予算消費も予定額より少なくなった。実際には、ゲノム編集用mRNAの調製の予備的検討として、レポーター遺伝子であるルシフェラーゼmRNAの調製を行った。また、肝実質細胞特異的非ウイルスベクターとして、ラクトース及びPEG修飾PAMAMデンドリマー(G4)の調製を試みた。2019年8月29日~9月1日に中国の成都市で開催された第10回アジアシクロデキストリンシンポジウムにて研究内容の一部を発表した。2020年度は、新型コロナウイルス感染拡大状況に依存するところもあるが、可能な限り研究の遅れを少しでも取り戻すべく尽力したい。, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Challenging Research (Exploratory), 2019/06 -2022/03, Grant-in-Aid for Challenging Research (Exploratory), Daiichi University, College of Pharmaceutical Sciences
    • Study on the role of CO as a signaling molecule in cell differentiation

      In this study, to understand the physiological function of carbon monoxide as an intracellular signal molecule, we investigated utilization of the artificial hemoglobin model complex, hemoCD. hemoCD is a supramolecular complex functions as an in vivo CO scavenger. First, the function of hemoCD on cells was examined. As a result, we have established an assay that accurately quantifies intracellular CO using hemoCD. This assay could also be applied to quantify CO in animal tissues, which has filed a patent application. Using this assay, the pharmacokinetics of CO was clarified.     • High-Performance Catalyst and Device Prepared by Metalloporphyrin

      本研究では、ヘム酸素の活性中心や、生体内での電子移動や酸素分子の運搬・貯蔵をつかさどる補因子のヘム分子(鉄ポルフィリン)の機能に着目し、鉄ポルフィリンに類似した金属ポルフィリン誘導体を基盤とする触媒およびデバイスの創製について、日本の研究グループとフランスの研究グループの各々の分子設計、合成、評価技術を結集して実施する。特に、ポルフィリン分子の骨格変換やシクロデキストリン等を用いた周辺の精密な分子環境設計を施し、金属ポルフィリン類縁体の新規触媒としての利用および、金属ポルフィリン類縁体の集積化に基づくエネルギー・電子移動デバイスの開発を行う。将来的には、分子レベルでの合理的な設計に基づく太陽電池や燃料電池の電極、人工光合成デバイスとして展開可能な基盤技術の獲得をめざす。 当該年度は、日本側の研究者とフランス側の共同研究者の間で、より密な研究討議を行い、それぞれが得意とするポルフィリン・ヘムタンパク質の素材合成を実施した。研究討議のために研究代表者である林はフランスのストラスブールで活動する共同研究者を11月初旬に訪問し、直接研究討議を行い、今後の方針を立てた。またフランス側が得意とするストラップ部位を導入したポルフィリンについて、日本側が得意とする水中でシクロデキストリンと複合化するための新たなデザインを導入した新規ストラップポルフィリン分子を設計し、その結果を論文として報告した。さらに、光捕集系をめざしたヘム蛋白質集合体の設計・合成にも着手し、幾つかのヘムタンパク質の規則的な複合体の構築と、複合体内での光駆動型のエネルギー移動について、予備的な結果も得た。, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Fund for the Promotion of Joint International Research (Fostering Joint International Research (B)), 2018/10 -2021/03, Fund for the Promotion of Joint International Research (Fostering Joint International Research (B)), Osaka University
    • Study on the role of endogenous CO in the circadian clock system

      In this study, we investigated the effect of endogenous CO depletion on the circadian clock system using highly selective CO scavenging molecule, hemoCD. Upon injection of hemoCD to mice, the expression levels of clock genes such as Per and Cry were significantly disrupted as a result of CO-depletion. The mechanistic study revealed the activity changes of CO-responsive transcriptional factors, CLOCK and NPAS2. In addition, CO-removal-induced inflammatory responses also caused the disruption of the clock genes. Our study clearly demonstrates, for the first time, that the depletion of endogenous CO induces disruption of circadian clock system.     • Experimental approach to respiratory and circulatory failure using artificial oxygen carriers and their derivatives

      Hemoglobin-based Oxygen carriers (HBOCs) have been studied to develop alternatives for red blood cell (RBC) transfusion. However, there is no single HBOC that is realized by now. Based on unique physical properties different from RBC, such as nanometer particle size (100-300 nm) and changeable oxygen affinity (P50O2 5-50mmHg), we tested available HBOCs in experimental setting of ischemia/reperfusion in partial or whole brain, myocardium, cochlea and skin ulcer. Accelerated healing of skin ulcer, stomach and bronchus were also noted. Sensitization of cancer radiotherapy or chemotherapy was observed. The mechanisms of action are considered to include, A) ubiquitous O2 as well as CO delivery, B) reduced oxidative stress, C) anti-inflammatory characteristics, D) anti-apoptotic property, resulting in E) preservation of organ/tissue function and morphology at the end.     • A stusy on biological function of endogenous CO as a byproduct of heme metabolism

      Carbon monoxide (CO) is continuously produced in mammalian cells during heme metabolic reaction. In this study, we investigated the biological function of the endogenous CO using a selective CO removal agent hemoCD. In order to introduce hemoCD into the cells, we conjugated hemoCD with octaarginine peptide. The new hemoCD derivative (R8-hemoCD) was taken by living cells and captured CO in cells, in which reactive oxygen species level was significantly enhanced in the CO-removed cells.     • Investigation of the role of intracellular CO using a supramolecular heme protein model complex showing high CO affinity

      Carbon monoxide (CO), a well known toxic gas, is continuously produced in the living body. Although the endogenous CO is considered to function in the biological system, the role of CO in biology has not been fully understood. Here, to investigate the biological function of endogenous CO, we utilized a supramolecular heme protein model complex, hemoCD, to remove endogenous CO from the living organisms. HemoCD was capable of quantitatively removing endogenous CO in the mice body due to its extremely high CO affinity. As a result, we succeeded to completely remove CO from the mice body, in which we found a biological feedback mechanism of endogenous CO to compensate the lack of Co in the living organisms (J. Am.     • Creation of a Supramolecular CO Receptor for Investigation of Biological Roles of Endogenously-Produced Internal CO

      Carbon monoxide (CO) is continuously produced in the eukaryotic organisms during degradation of hemin by heme oxygenase (HO). The biological roles of CO in the living organisms have not been fully understood because of lacking of knockdown method for endogenously produced CO. We have shown that a supramolecular complex, hemoCD, strongly binds CO higher than that of hemoglobin. In the present study, hemoCD was used for removal of CO in the living organisms and genetic response upon removal of CO was investigated. Furthermore, for monitoring the depletion effect of CO in the human cells, the hemoCD scaffold was conjugated with an octaarginine peptide that has a strong ability to bring molecules into the cell interior.     • An Artifi cial Hemoglobin Drug for Toxic Gases Generated in the Fire Accidents

      Summary: The most toxic gases during combustioon are carbon monoxide (CO) and hydrogen cyanide (HCN). When these two types of gas are inhaled at the same time, they are extremely toxic. In this study, we developed a drug that can remove CO and HCN at the same time.
      Usage Apply: Emergency medicine at the fire accident place, CO/HCN remover for improving sequelae after the gas poisoning.
      Remarks: In many cases, HCN and CO are poisoned at the same time in a fi re accident, and there is no method to removeboth at the same time. In particular, HCN poisoning in fi re is not as socially recognized as CO poisoning, despite itshigh lethality. This drug is cost eff ective, stable and convenient, and can remove CO and HCN in a single dose.

      Summary: Whereas carbon monoxide (CO) is well known as a toxic gas, CO is continuously produced in our body. CO works as a signal messenger in the living systems, mainly working for cell-protection. In our laboratory, we have been developing a selective CO removal and delivery agents, which are useful for life science research to elucidate the biological function of CO. Similar but different from nitric oxide (NO), CO exerts an unique biological properties and therefore we can use CO as a medicinal tool.
