  • Precise Formation of Catalysts Having Specified Fields through Fixation of Metal Complexes in Biological Molecules

    水中においても安定なカルボキサミドの鉄イオンへの配位に着目し、過酸化水素を酸化剤として選択的にアルカンを酸化する単核非ヘム鉄錯体Fe(dpaq)を開発してきた。Fe(dpaq)は金属酵素の機能の一部を再現するが、反応速度、基質特異性の面で天然酵素に及ばない。そのため、この錯体触媒をもとに高性能な人工酵素の創製を目指した。Fe(dpaq)錯体はヒト血清アルブミンの薬物結合サイトに強く結合する。種々の条件において酸化触媒能を検討した結果、Fe(dpaq)アルブミン複合体は過酸化水素を酸化剤としてプラスミドDNAを効率よく切断することを発見した。この結果は、Fe(dpaq)は蛋白質内部においても酸化触媒活性を発現できることを実証するものである。この結果をもとに、アルブミンの疎水ポケットに結合した鉄アミド錯体による不斉スルホキシド化を検討した。触媒回転数は、アセトフェノンを内部基準としGC分析により行い、不斉選択性はキラルカラムによるHPLC分析により行った。用いた鉄アミド錯体は、有機溶媒中で過酸化水素を用いた優れた選択アルカン酸化触媒となることが判明しているFe(dpaq)、水中で過酸化水素による一電子酸化反応活性(ペルオキシダーゼ活性)を示すことが判明している窒素4座カルボキシルアミド鉄錯体Fe(mpaq)、さらに、L-プロリンを出発原料とする窒素4座カルボキシルアミド鉄錯体Fe(Propaq)である。Fe(dpaq)は不斉を持たず、Fe(mpaq)はラセミ体として存在するが、Fe(Propaq)は不斉錯体である。不斉鉄錯体を用いた先行研究の反応条件に従い、触媒反応を評価した結果、錯体のみでは触媒回転数も低く、不斉選択性が観測されないが、アルブミン存在下では、触媒回転数が向上すること(Fe(dpaq)では10倍以上)、不斉選択性も僅かであるが観測されることが判明した。, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Scientific Research on Innovative Areas (Research in a proposed research area), 2018/04 -2020/03, Grant-in-Aid for Scientific Research on Innovative Areas (Research in a proposed research area), Doshisha University

  • Development of Small-molecule Fluorescent Probes Based on the Reactivity of Metal Complexes

    We have developed a fluorescent probe that can rapidly visualize hydrogen peroxide released from cells as a signal. Fluorescent probes comprise complexes, profluorophores, and linkers linking them. In this study, the complex structure, profluorophore, and linker were improved. Hydrogen peroxide could be detected even in HeLa cells which do not overexpress the EGF receptor, when the most sensitive hydrogen peroxide fluorescent probe was used. In addition, it was shown that the emission of nitrogen monoxide could be achieved in the hypoxia region of cell spheroids by using MnNO complex.     鉄錯体を触媒とするアルカンのC-H結合選択的な水酸化反応が報告されており、その有機合成化学への利用が期待されている。これらの触媒を用いれば、過酸化水素によって、基質の中の最も酸化されやすいC-H結合を高い選択性をもって触媒的に酸化することができるが、有機合成の現場で酸化したいC-H結合と最も酸化されやすいC-H結合とが一致するケースは多くない。本研究では、基質の酸化されやすさに依存しないC-H結合の酸化のための精密制御反応場を鉄錯体の近傍に構築し、従来では不可能であった位置にあるC-H結合の選択的酸化反応を可能にすることを目指した。また、鉄錯体を蛋白質とハイブリッド化し、水中で酵素型触媒反応を実現することも目指した。ポルフィリン分子、さらに、ベンゼン環を5つ有するイプチセン誘導体を極めて嵩高い立体障害基として有する窒素配位子を合成することに成功した。また、A03班の大阪大学林高史教授、小野田晃准教授との共同研究によって、ヒト血清アルブミンの疎水ポケットに我々が保有する鉄錯体の一つFe(dpaq)が特異的に結合することが判明した。水中において鉄錯体-蛋白質ハイブリッドが酸化活性を示すか、酸化活性を調べた結果、過酸化水素によるプラスミドDNA酸化的切断に高い活性を示すことを見出した。Fe(dpaq)と比較した場合、その酸化活性は約2倍となった。この結果は、蛋白質とDNAとの相互作用によって、DNA酸化活性が向上したと考えられた。Fe(dpaq)に生体ポリアミンとして知られるスペルミンを結合させた場合にも、プラスミドDNAの酸化的切断が向上することを見出した。さらに、配位子に一連の置換基を導入し、鉄錯体の酸化還元電位とDNA酸化切断活性の相関を調べた結果、DNA酸化反応の初速の対数値と酸化還元電位に直線相関関係があり、鉄中心が還元されやすくなるほど活性が高くなることが分かった。, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Scientific Research on Innovative Areas (Research in a proposed research area), 2016/04 -2018/03, Grant-in-Aid for Scientific Research on Innovative Areas (Research in a proposed research area), Doshisha University

    Inspired by the reaction mechanism of mononuclear nonheme iron enzyme, SyrB2, which catalyzes direct halogenation of alkane C-H bonds under mild conditions, novel fluorination catalysts have been designed. SyrB2 generates nonheme oxoiron(IV) species as an active oxidant, which has a halogen ligand coordinated cis to the oxygen ligand. In order to reproduce the biological oxidant, new iron complexes that contain two cis-labile coordination sites have been synthesized.     鉄オキソ種を活性種とする触媒によるLate-stage Oxidationのための選択酸化反応が脚光を浴びている。しかし、これらの触媒が真に有機合成の現場で使用されるには、触媒回転数が低い、選択性が基質の酸化されやすさに依存している等の克服すべき課題がある。金属酵素の酸化活性種となる金属オキソ種は、対応するパーオキソ種のO-O結合の開裂により生成する。本研究では、金属酵素が行っているパーオキソ種のO-O結合の開裂により生成する金属オキソ種への変換を触媒設計に取り入れる。具体的には、鉄パーオキソ種から鉄オキソ種を行い、かつ基質結合によって鉄オキソ種を生成する鉄錯体を合成し、高い触媒回転数と位置特異的酸化を達成する。これまでに、過酸化水素によるアルカンの水酸化反応に対して例外的に高い選択性と耐久性を示す鉄錯体触媒を開発してきた。また、触媒活性の向上だけでなく、触媒活性種である鉄オキソ種の単離と反応解析も行ってきた。本研究では、カルボン酸の添加によって、過酸化水素の酸素―酸素結合の開裂が誘起され、鉄オキソ種の生成が促進されることを基質酸化および吸収スペクトル変化より確認することに成功した。さらに、カルボン酸を有する炭化水素を基質として用いた場合には、カルボン酸部位が配向基として働くために、位置特異的な酸化反応が進行することも見いだした。種々のカルボン酸を用いてアルカン酸化の選択性を評価した結果、立体障害の大きなカルボン酸を用いた場合、選択性を低下させずに触媒回転数が向上することを見いだした。, 日本学術振興会, 科学研究費助成事業 新学術領域研究(研究領域提案型), 2015/04 -2017/03, 新学術領域研究(研究領域提案型), 同志社大学

  • Synthesis of Metal Complexes that can function in an Aqueous Solution Aiming at Application to Chemical Biology

    For the purpose of developing bio-inspired metal complexes applicable to chemical biology, we synthesized metal complexes with newly designed various ligands that specifically stabilize the structures of the metal complexes in solution. With the metal complexes, we successfully attained following biologically relevant functions, (1) mimicking P-to-Q conversion in the O_2-activation of soluble methane monooxygenase(sMMO), (2) roles of the carboxylate-rich coordination environment in O_2-activating diiron enzymes, and (3) enhancement of catalytic activity of ruthenium complexes for O_2-evolution by water oxidation as a model of oxygen evolution complex(OEC).   • Selective synthesis of luminescent metal nanoclusters for intracellular imaging

    A new porphyrin-based passivant was synthesized which has two disulfide bonds in the same direction. Small gold nanoclusters were easily obtained when gold(III) ion was reduced in the presence of the passivants. Resulting gold nanoclusters exhibited an intense fluorescence due to the porphyrin molecules.     アミドアニオン配位を有する単核鉄錯体を開発し,過酸化水素およびアルキルペルオキシドとの反応を解析した.その結果,ヒドロペルキソ付加種の生成には,塩基の共存が必須であるが,この塩基の塩基性の程度がヒドロペルキソ種の安定性に影響を与えることをみいだした.この結果は,過酸化水素からプロトンを受け取った塩基が,ヒドロペルキソ種に対して共役酸として働き,ヒドロペルキソ種の酸素-酸素結合のヘテロリシスを誘起した結果であることを強く示唆している.そこで,pKaの様々に異なる一連の塩基を用いて,ヒドロペルキソ種の安定性を評価し,添加した塩基の酸塩基触媒としての機能を評価し,共役酸の酸性度とヒドロペルオキソ種との安定性の間に良い相関関係が存在することを明らかにした.また,共役酸はペルオキシド中間体の安定性を低下させるだけでなく,酸素-素結合のイオン的開裂を促進することも明らかにすることができた.これらは,金属酵素の反応中心で金属イオンに配位しないアミノ酸側鎖が第2配位圏で行う協奏機能を人工系において発現可能であることを実証した点で重要である.本研究では,本金属錯体が水中でも安定に存在することに着眼し,水中での過酸化水素の活性化および外部基質の酸化へと展開した.その結果,上記の5座配位金属錯体ではなく,4座モノアミドアニオン配位単核鉄錯体が高いペルオキシダーゼ活性を発現することを見いだした.反応解析の結果,新しく開発した鉄錯体には空配位座がシスの関係に存在しているために,過酸化水素の活性化においても,基質酸化においても,その反応速度定数が向上していることが判明した., 日本学術振興会, 科学研究費助成事業 特定領域研究, 2008 -2009, 特定領域研究, 同志社大学
  • Development of highly efficient reaction process for aerobic selective oxidation on the basis of photocatalytic conversion

    In the vapor phase selective photocatalytic oxidation of alkanes, alkali-ion-modified silica-supported vanadium oxide was found to be appropriate catalyst system while in the liquid phase selective photocatalytic oxidation, alumina-supported vanadium oxide was found to be suitable. The general mechanism of photocatalytic oxidation of alkanes was proposed. In the liquid phase selective photocatalytic oxidation of alcohols, niobium oxide itself was found to be the best and its reaction mechanism was proposed. Although low-loaded vanadium oxide supported on silica was known to be effective for photo-epoxidation of propene with molecular oxygen, the active species is now under discussion. New photocatalytic system with porphyrin complex was developed for liquid phase photocatalytic oxidation of alkenes.     生体内において、選択的な酸化反応を行う金属酵素、例えば,ヘム依存性のHorseradish Peroxidase(HRP)は、アミノ酸によるPush-Pull効果によって、ヘテロリシスを誘起することが知られている。この点に着眼し、強いドナー性配位となるアミドアニオン配位を導入した単核鉄錯体を設計、合成した。本錯体は、マイナス40度の低温下、過酸化水素との反応により、単核鉄3価ヒドロペルオキシド種を与えた。安定性を評価した結果、Push効果によって、ヒドロペルオキシド基の酸素-酸素結合のヘテロリシスの誘起されることを見いだした。また、シクロヘキセンを酸化基質とし、触媒酸化反応を検討した結果、ドナー性の高いアミドアニオン配位子を用いるほど、鉄3価ヒドロペルオキシド種の安定性が低下することを見出した。また、中性の配位子を用いた場合のように酸素-酸素結合のホモリシスが優先的に進行するのではなく、テロリシスが誘起されるために、シクロヘキセンのアリル位の水酸化ではなく、エポキシ化生成物が得られることが判明した。この結果は、アミドアニオン配位の導入によるPush効果によって、単核鉄3価ヒドロペルオキシド種の酸素-酸素結合のヘテロリシスが進行し、アリル位の酸化の原因となるヒドロキシルラジカルの生成が抑止されたためと考えられる。, 日本学術振興会, 科学研究費助成事業 特定領域研究, 2007 -2007, 特定領域研究, 京都大学
  • ポルフィリン金属錯体による分子表面認識とナノサイズ反応場の構築

    十分に設計された配位子を用いれば、生成される金ナノ粒子の粒子径だけでなく、その形状、電子状態、反応性の制御、反応空間の構築も可能であり、その先に豊かな金ナノクラスター科学を展開できるのではないかという着想の基に、『ポルフィリン金属錯体による分子表面認識とナノサイズ反応場の構築』を目指して、金ナノ粒子表面を認識する強固に認識する表面保護配位子Por_ClSを設計、合成した。本表面保護配位子Por_ClSは、ポルフィリン骨格から同一方向に4つのアセチル基によって保護された硫黄原子を提示しており、金表面を認識することが可能である。当初の分子設計通り、表面保護配位子Por_ClSの存在下、HAuCl4を還元することで、Por_ClSによって表面が保護された極めて安定な金ナノクラスターが調製可能であることを見出した。また、ポルフィリン表面保護配位子のHAuCl4に対する仕込み比を増加させることによって、粒子径分布の狭い金ナノクラスターを調製することにも成功した(2.0±0.2nm)。この知見を基に、アセチル基によって保護された硫黄原子ではなく、ジスルフィド結合を有するポルフィリン表面保護配位子を設計、合成した結果、ポルフィリン表面保護配位子のHAuCl4に対する仕込み比に関係なく、粒子径分布の狭い金ナノクラスターが得られることを見いだした。また、得られたポルフィリン保護金ナノクラスターが自己組織化し、超格子を形成することも判明した。, 日本学術振興会, 科学研究費助成事業 若手研究(B), 2005 -2007, 若手研究(B), 京都大学
  • Development of low temperature processes of deNOx with ammonia and oxidative removal of ammonia

    The aim of the resent research is the development of low temperature selective catalytic reduction of nitrogen oxides with ammonia using photo-energy (photo-SCO) and the goal is the development of low temperature selective catalytic oxidation of ammonia on the basis of photo-SCO. We had found that NO can be reduced selectively by ammonia over photoirradiated titania in the presence of excess oxygen. During the first two years, the catalyst composition was optimized and the catalyst system operated under sunlight and fluorescent light was developed. Through this study, we could clarify the detailed reaction mechanism. Catalytic activity depends on the phase of titania but the response to the visible light does not. This result was deduced by observation of electron transfer from amide anion, an intermediated, to titanium cation 3d orbital with ESR and action spectrum. As an additive component to titania, molybdenum, niobium, and tungsten were found to be effective. The modification of titania with these metal cations brings about the enhancement of the acid strength of titania, resulting in the acceleration of the rate-determining step. During the last two years, we developed the catalytic system for abatement of ammonia at low temperature by applying the photo-SCR. Kinetic analysis of reaction mechanism and the infrared study led us to the finding that TiO-11 is the most effective catalyst.     本研究では合目的配位子デザインにより人工非ヘム鉄酵素の構築を目指した。研究の前半では、鉄3価イオンを活性中心に有し、分子状酸素を用いてカテコールを開裂する非ヘム鉄酵素であるカテコールジオキシゲナーゼに着目した。(1)新規水溶性非ヘム鉄錯体を用いて水溶液中におけるカテコールの酸素添加開裂反応を可能にし、pH依存性等の反応機構に関わる因子を明らかにした。(2)種々の置換基を有するカテコール配位子から一連の非ヘム鉄錯体を調製し、電子状態に対するカテコール配位子上の置換基効果を詳細に検討した。これらの錯体と酸素分子との反応速度の対数が、カテコールから鉄(III)イオンへの電荷移動エネルギーと良好な直線関係を示すことを初めて明らかにした。この結果を基に、鉄2価セミキノン励起状態が関与し、鉄3価イオン上で酸素分子が活性化される新しいカテコールジオキシゲナーゼの反応機構を提唱した。(3)一方、電子吸引性基を有する窒素4座配位子を有する錯体では低スピン状態が誘起されることが判明した。これらの低スピン錯体種は高スピン錯体種に比べ著しく反応性が低いことが明らかとなった。(4)鉄3価イオン上で酸素分子が活性化を促進するため、空配位座を有する鉄3価カテコール錯体をデザインしたところ、酸素分子と迅速に反応するだけでなく、比較的安定な酸素錯体を形成することが明らかとなった。更に、酸素錯体の形成が可逆過程であることを明らかにした。(5)一方、マンガン2価セミキノン錯体は分子状酸素と反応し、定量的にキノンとマンガン2核(III,III)ビス-mu-オキソ錯体を形成した。また、この2核マンガン錯体の溶液にカテコールを加えたところ、マンガン2価セミキノン錯体が再生した。これらを基に、高効率のカテコール酸化触媒システムを構築することに成功した。反応機構を詳細に検討した結果、本システムは従来のカテコール酸化反応と異なり、分子状酸素を水に還元することが判明した。, 日本学術振興会, 科学研究費助成事業 若手研究(B), 2002 -2004, 若手研究(B), 京都大学

  • Development of ammonia SCR system with self-assembled photocatalyst

    We have synthesized supported metal oxide catalysts by self-assembling of the active composites and evaluate the photocatalytic activity of thus prepared catalysts. To evaluate the capability of the catalysts for partial oxidations, we carried out photooxidation of light alkanes and liquid phase photooxidation of cyclohexane. We found that Rb2O-V2O5/SiO2 exhibited high activity for the former reaction and response to visible light while for the latter V2O5/Al2O3 catalyst was found to be effective. Titanium dioxide showed comparable high activity but catalyze non selective oxidation. We carried out the evaluation of these catalyst candidates for low temperature ammonia SCR reaction (NO photo-SCR with ammonia). The reaction was carried out in a continuous flow reactor with a fixed catalyst bed which models a practical condition. The practical condition means the feed, GHSV = 4000 h-1, NO and ammonia -1000 ppm, O2 2%. Opposite to the results of partial oxidation, V2O5/Al2O3 was almost inactive and Rb2O-V2O5/SiO2 exhibited 40 % conversion of NO while TiO2 exhibited the highest activity 80% conversion rate was maintained for 100 h. We speculated that ammonia SCR is fundamentally equal to the total oxidation. By varying the reaction conditions and spectral consideration with FT-IR, UV/VIS, and ESR, we proposed the reaction mechanism of a series of elemental steps (1) adsorption of ammonia onto titanium oxide, (2) photoactivation of adsorbed ammonia, (3) reduction of NO as well as the reduction of Ti cations and (4) re-oxidation of reduced Ti cations. In the absence of oxygen. NO acts as an oxidant of the reduced Ti cations.   • Selecteve catalytic reduction of NO with CO in the presence of oxygen

    Since an outbreak of Nox gas in high temperature burning is inevitable, the reduction and/or removal of Nox is strongly requested from the viewpoint of environmental preservation. Although there have been a series of studies aiming at reduction and/or removal of Nox exhausted from diesel engine and lean-burn engine by reduction with hydrocarbons as an reductant, yet any catalysis system has not been used in practice because of the dependence of activity upon the kind of hydrocarbon and because of the catalyst life. In the present study, we attempted to develop the Nox removal SCR system with CO as a reductant, Cu/Al203 exhibits as high catalytic activity in the presence of oxygen as Rh/A1203, a model three-way catalyst, and higher activity in the region of lean-burn than Rh/A1203. In the term of project, we have found out the correlation between catalyst state and activity in the lean-burn condition and carried out the optimization of the catalyst. Catalytic activity varied with the loading of Cu and 1 wt% loaded catalyst exhibited the highest activity. But 0.25 wt% loaded catalyst exhibited highest TOF (turn-over-frequency) . ESR and XAFS showed that Cu species which is sintered by high temperature treatment exhibits high activity for CO oxidation resulting in the prevention from NO reduction. The Cu species effective for NO reduction was found to be highly dispersed mononuclear Cu species which is dominant species in 0.25 wt% loaded catalyst TPR spectral features of this catalyst showed that this Cu species is hardly reduced and the H2 reduction at higher than 1000 K does not bring about the formation of metallic Cu but monovalent Cu.   • Studies on development of the selective oxygenation catalysts based on mimicking function of nonheme monoiron oxygenases

    Following studies were performed on the development of oxygenase-like catalytic functions by nonheme monoiron complexes and manganese complexes and on the clarification of mechanisms of electron transfer that is essential for activation of substrates. 1)Intradiol and extradiol oxygenative cleavage of catechols by monoiron complexes Catalytic activity of oxygenations by monoiron complexes ligated by tripodal ligands and tridentate ligands were studied. Effect of electronic and steric effects of substituents on the selective intradiol cleavage gave important information of the oxygen activation and oxygen insertion process into the C-C bonds. In the extraiol cleavage by iron complexes with tridentate ligands also clarified the mechanism of oxygenation via the direct oxygen binding to the iron center. Not only ligands with only the N binding sites but also ligands with N and O binding sites were used for clarification of the O2 insertion into catechols. 2)Intra and extradiol oxygenative cleavage of catechols by manganese complexes By mimicking manganese-containing extradiol cleaving oxygenases, Mn(II)-semiquinone complexes were first synthesized. The complexes were found to be oxygenated to give the intradiol cleavage productws. Interestingly, the reversible uptake of oxygen was observed, indicating that the molecular oxygen binds first to the metal center (iron and manganese) prior to binding to the catechol carbon. 3)Oxygenations in water by iron complexes with water-soluble ligands Although enzymatic oxygenations proceeds in aqueous media, models oxygenations have been performed in organic solvents. This is partly due to the ligands soluble only in organic solvents, and partly on exclusion of oxygen saurce other than molecular oxygen. We have first developed a model system that works in water to give the selective intradiol cleavage of mono-alkyl catechols, e.g. 4-butyl catechols, catechols with the electron-withdrawing substituents, such as chloro and carboxylate, by using the TPA ligands with sulfonate subsitutients. It is important to find that the protocatechuic acid was found to be oxygenatively cleaved by monoiron complexes, because the model oxygenation has not be developed in spite of the most popular enzyme, protocatechuic oxygenases. It was also found that the pH control of the media is essential for the aqueous system.
