Research Projects

  • Reflection and adsorption of low energy hydrogen at solid surface

    同志社大学に小型の実験装置を新規設営するため,R5年度は新装置設計期間とし,年度中の実験データ採取には核融合科学研究所に設置した装置を用いた.モリブデン(Mo)とC12A7(12CaO-7Al_2O_3)エレクトライド材料を用いた実験を行い,得られたデータの解析を行った結果,表面粗さに起因する角度分布の変化と,表面仕事関数の大きさに依存した正イオン/負イオン比率の変化が観測されることが明らかとなった.結果の一部をまとめ,Journal論文原稿として提出した. 加速器用負イオン源実機に現れる表面生成の効果を調査するため,J-PARCイオン源研究チームと共同研究を行い,高速エミッタンスメーターを開発してこれを使用し,表面生成に起因して発生する水素負イオンビームの空間分布時間発展を調査した.実験結果はシミュレーション計算結果と比較・検討した.内容を国際イオン源会議に発表し,論文(会議抄録)にまとめた. 設計中の小型装置主要部分となる磁気四重極型質量分離機/低エネルギービーム収束系については,簡易計算による概念設計を終え,三次元磁場計算に基づく詳細設計に入った.低エネルギーイオン源については静電レンズ系を取り去り,引き出し電極構造により磁気レンズ系に合わせる構造とするため,軸対象系のビーム引き出しシミュレーションが行えるまでの,プログラム開発を行った. 今年度の実験に用いた核融合科学研究所に整備した大型装置では,入射角の精度を高める目的から入射イオンの飛行距離を数mとしている.低エネルギービームの空間電荷による発散のため,本大型装置を用いた実験ではビームエネルギーを下げることが困難である.そこで小型の,90度磁場偏向型質量分析器を備えたイオン源を核融合研に設置した装置に取り付け,その性能評価を行ったところ,十分な強度・分解能が得られないことが分かった., Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, 2023/04 -2026/03, Grant-in-Aid for Scientific Research (B), Doshisha University
  • Multiscale Theories of Genome Modality

    SMCタンパク質のDNAループ押し出しの分子機構を解明するために、残基粒度を持つ粗視化分子動力学シミュレーションによって、バクテリアSMC蛋白質、およびコヒーシンのDNA結合様式を研究した。バクテリアScpA-SMC蛋白質のATP加水分解依存的な構造ダイナミクスを再現し、ATP型Engaged構造、ADP型O構造、Apo型Disangaged構造それぞれの前兆モデルを得た。それをもとにその各々のDNA結合様式を明らかにした。 ゲノムDNA高次構造転移が示す不連続性(all-or-none)によって誘起される遺伝子発現活性の促進と抑制の二相性について、対イオン凝縮理論に対イオンの並進エントロピー効果を取り入れた理論モデルを構築すると共に、蛍光顕微鏡によるDNA一分子計測、無細胞発現系実験を実施することで、DNA高次構造転移が引き起こす遺伝子発現ON/OFFスイッチング機構メカニズムを明らかにする。 染色体マルチスケール理論の一端を担うメゾスケール高分子モデルの構築を進めた。具体的には、ヌクレオソームを球形粒子、リンカーDNAを非線形バネとした粗視化高分子モデルをベースにブラウン運動を考察し、ヒト細胞核内において多発するヌクレオソーム間衝突に着目して、衝突時の跳ね返りを含むブラウン動力学アルゴリズムを考察、実装した。メゾスケール動力学の構築に重要かつ必須なステップとなった。 コヒーシンのループ押し出し運動と転写因子/RNA合成酵素による遺伝子とエンハンサの転写凝集体への結合・解離ダイナミクスを考慮に入れて 、転写凝集体表面のクロマチンの運動と転写ダイナミクスの解析を行った。遺伝子-エンハンサ間のDNAの長さを長くすると緩和時間が長くなるため、次のループ押し出しが始まるまで遺伝子が凝集体表面付近に留まるので、遺伝子の凝集体内の転写装置へのアクセス率が高くなることが分かった。, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Transformative Research Areas (A), 2020/11 -2025/03, Coinvestigator, Grant-in-Aid for Transformative Research Areas (A), Kyoto University

  • Contribution of higher-order structure transition of giant DNA on gene expression efficiency

    I have studied a contribution for gene expression efficiency by higher-order structure transition of giant DNA. I have found that an enhancement of gene expression efficiency result in a conformation of giant DNA just before globule state. This mean that the enhancement is due to the parallel ordering of DNA segments. Meanwhile, an inhibition on gene expression is caused by the compaction of DNA into a tightly packed state.   • Observation by infrared spectroscopy on time-development growth of carbon dusts in hydrogen plasma

    We have studied the formation processes of carbon dust using the small experimental setup which we have been designed and built. The micron sized carbon dusts have been observed in the designed experimental setup after hydrogen plasma discharge. Almost of all carbon dusts have a spherical shape which are of the order of micro meter. The images of cross sections of carbon dusts observed by an electron scanning microscopy have indicated that the carbon dusts growth by columnar growth. The infrared spectroscopy have been adopted to observe the dynamic formation of carbon dusts. The experimental results have indicated that spectra of infrared spectroscopy have large amount of noise.   • Development of Lead-Free Biocompatible Multi-Layer Thin-film Structure Generation Technology for Implantable Actuator and Energy Harvest Device

    We developed a lead-free biocompatible multi-layer thin-film structure generation technology for implantable actuator and energy harvest device, as demonstrated below: (1) Piezoelectric stress constants of designed mixed crystals, Ti doped MSO, obtained by the first-principle calculations was 0.696(C/m2), which is 1.52 times larger than MSO. Piezoelectric constant d33 was 354.8pm/V. (2) Optimum post-annealing temperature was obtained as 676 centigrade, its crystallization of tetragonal MgSiO3 was 90.5%.High crystallization of tetragonal MgSiO3 was obtained on generated ten-layered thin film. Its piezoelectric constant d33 was 406.1 pm/V. (3) Mono-morph MSO cantilever actuator could be vibrated with the amplitude tens nm under 15 Voltage. (4) A magnetic induced hybrid vibration energy harvester, which employed the mono-morph MSO and PVDF piezoelectric cantilever, generated 1.2 microW.   • Theory and code development for evaluation of tritium retention and exhaust in fusion reactor

    Through a cooperative research on code development by domestic and overseas researchers for divertor/SOL plasma physics and tritium plasma wall interaction, an integrated simulation code was developed in order to evaluate accurately tritium retention and exhaust in fusion reactors. It includes development of each physical models by means of cross-code and code-experiment comparisons in laboratory experiments and validation and improvement of the integrated code by analyzing experiments in present fusion devices.   • Novel method for constructing a stable 3D cellular assembly by near-infrared laser

    Summary: We introduce a novel method for constructing stable 3D cellular assemblies by using optical tweezers with dextran. One of advantages of this method is that it constructs a stable cell to cell contact within a few minutes. Our newly established method is expected to be useful for next-generation 3D single-cell assembly systems in the field of regenerative medicine and tissue engineering.
    So far, artificial scaffolds have often been used to construct 3D cellular assemblies. However, scaffolds used for cellular assemblies are sometimes toxic and may change the properties of the cells. On the contrary, this technology enabled us to construct a 3D cellular organism under conditions close to the in vivo cellular environment.
